Die Verrottung der Körner durch Samen-Pilzresistenz stellt eine der größten wirtschaftlichen Herausforderungen zu Getreideproduktion weltweit, nicht zu ernsthaften Gefahren für die menschliche und tierische Gesundheit zu erwähnen. Unter Getreideproduktion ist Mais wohl die betroffenen Kultur, durch Pathogen-induzierte Verluste in Korn Integrität und Mykotoxin-Verunreinigung von Saatgut. Die beiden am weitesten verbreiteten und problematisch für Mykotoxine Maiserzeuger und Lebens-und Futtermittel-Prozessoren sind Aflatoxin und Fumonisin, durch Aspergillus flavus und Fusarium verticillioides produziert bzw.. Neuere Untersuchungen in der molekularen Pflanze-Pathogen-Interaktionen haben Versprechen für das Verständnis spezifischer Mechanismen mit pflanzlichen Reaktionen auf Pilzinfektion und Mykotoxinkontamination 1,2,3,4,5,6 verbunden demonstriert. Da viele Labs verwenden Kernel-Assays, um Pflanzen-Pathogen-Interaktionen zu untersuchen, besteht ein Bedarf für ein standardisiertes Verfahren zur Quantifizierung von unterschiedlichen biologischen Parameter, soErgebnisse aus verschiedenen Laboratorien können Cross-interpretiert werden. Für eine robuste und reproduzierbare Mittel für quantitative Analysen auf Saatgut, haben wir im Labor Kernel-Assays und anschließender Methoden entwickelt, um das Pilzwachstum, Biomasse und Mykotoxinkontamination quantifizieren. Vier sterilisiert Maiskörner in Glasfläschchen mit einer Pilz-Suspension (10 6) beimpft und für einen vorbestimmten Zeitraum. Probenfläschchen werden dann für die Zählung von Conidien mit einem Hämocytometer, Ergosterin-Biomasse Analyse ausgewählt durch Hochleistungs-Flüssigchromatographie (HPLC), Aflatoxin Quantifizierung mit Hilfe eines AflaTest Fluorometer Verfahren und Fumonisin Quantifizierung durch HPLC.
