Het beenmerg is de belangrijkste site waar HSC&#39;s en meer volwassen bloedcellen afkomst voorlopercellen verblijven en te differentiëren in een volwassen organisme. HSC&#39;s vormen een minuut celpopulatie van pluripotente cellen die in staat het genereren van alle bloed-cellijnen voor een life-time 1. De moleculaire dissectie van HSC homeostase in het beenmerg heeft belangrijke gevolgen voor hematopoiese, oncologie en regeneratieve geneeskunde. We beschrijven de etikettering protocol met fluorescente antilichamen en de elektronische gating procedure in flowcytometrie om hematopoietische voorlopercellen subsets en HSC&#39;s in individuele muizen (fig. 1) te scoren. Daarnaast beschrijven we een methode om op grote schaal verrijken hematopoietische voorlopercellen als lange termijn (LT) en korte termijn (ST) reconstructie van HSC uit gepoold beenmerg celsuspensies door magnetische verrijking van cellen die c-Kit. De resulterende cel preparaat kan worden gebruikt om geselecteerde subsets sorteren van in vitro enin vivo studies functionele (Fig. 2). Beide trabeculaire osteoblasten 2,3 en sinusvormige endotheel 4 vormen functionele niches ondersteunen HSC&#39;s in het beenmerg. Een aantal mechanismen in de osteoblastische niche, met inbegrip van een deelverzameling van N-cadherine + osteoblasten 3 en interactie van de receptor tyrosine kinase Tie2 uitgedrukt in HSCs met zijn ligand angiopoietine-1 5 eens bij het ​​bepalen van HSC rust. &quot;Winterslaap&quot; in het beenmerg is cruciaal voor HSC&#39;s te beschermen tegen de replicatie en de uiteindelijke uitputting na overmatige fietsen activiteit 6. Exogene stimuli die op cellen van het aangeboren immuunsysteem, zoals Toll-like receptor liganden 7 en interferon-α 6 kan proliferatie en differentiatie van HSC ook leiden tot geslacht toegewijd voorouders. Onlangs heeft een bevolking van slapende muis HSCs binnen de lin - c-Kit + Sca-1 + CD150 + CD48 - CD34 - de bevolking is beschreven 8. Het sorteren van cellen op basis van CD34 expressie van de hematopoietische voorlopercellen verrijkte celsuspensie zoals hier beschreven kan de isolatie van zowel latente zelfvernieuwende LT-HSC&#39;s en ST-HSCs 9. Een soortgelijke procedure op depletie van lijn positieve cellen en sorteren van LT-HSC met CD48 en Flk2 antilichamen is eerder beschreven 10. In dit rapport geven we een protocol voor de fenotypische karakterisatie en ex vivo celcyclus analyse van hematopoietische voorlopercellen, die nuttig kan zijn voor het toezicht hematopoiese in verschillende fysiologische en pathologische omstandigheden. Bovendien beschrijven we een FACS sorteren procedure HSCs, die kunnen worden gebruikt om factoren en mechanismen reguleren van hun zelfvernieuwing expansie en differentiatie in celbiologie en signaaltransductie assays en voor transplantatie definiëren.
