Dynamische regulatie van de Rho familie van kleine guanosine triphosphatases (GTPases) met veel ruimte en tijd precisie essentieel is voor de verschillende cellulaire functies en gebeurtenissen 1, 2. Hun spatiotemporeel dynamische karakter is geopenbaard door de visualisatie van hun activiteit en lokalisatie in real-time 3. Om dieper inzicht in hun rol in diverse cellulaire functies op moleculair niveau te krijgen, moet verstoring van eiwit de activiteiten van de volgende stap zijn op een precieze subcellulaire locatie en timing. Om dit doel te bereiken, hebben we een methode ontwikkeld voor licht-geïnduceerde, spatio-temporeel gecontroleerde activatie van kleine GTPases door het combineren van twee technieken: (1) rapamycine-geïnduceerde FKBP-FRB heterodimerisatie en (2) een foto-kooien methode van rapamycine. Met het gebruik van rapamycine-gemedieerde FKBP-FRB heterodimerisatie, hebben we een methode ontwikkeld voor het snel induceerbaar activatie of inactivatie van kleine GTPases including Rac 4 Cdc42 4 RhoA 4 en Ras 5, waarbij rapamycine induceert translocatie van FKBP-gefuseerde GTPases of hun activatoren naar de plasmamembraan waar FRB verankerd. Voor de koppeling met deze heterodimerisatie systeem, hebben we ook een foto-kooien systeem van rapamycine analogen. Een foto-kooi verbinding is een klein molecuul waarvan de activiteit wordt onderdrukt met een photocleavable beschermende groep bekend als een kooi groep. Om heterodimerisatie activiteit volledig onderdrukken, hebben we een gekooide rapamycine dat gebonden is aan een macromolecuul dat de verkregen grote complex niet de plasmamembraan steken, waardoor vrijwel geen achtergrondactiviteit als chemisch dimerizer in cellen 6. Figuur 1 toont een schema van onze systeem. Met de combinatie van deze twee systemen, wij lokaal aangeworven a Rac activator aan de plasmamembraan op een tijdschaal van seconden en bereikt het licht-geïnduceerde Rac activatie aan de subcellulaire level 6.
