De kwaliteit van de MES-cellen is de meest kritische parameter voor een efficiënte productie van motorische neuronen. MES cellen moeten worden geteeld op PMEF om spontane differentiatie te voorkomen. Direct differentiatie van embryonale stamcellen (ES cellen) in functionele motorische neuronen een veelbelovende bron van de ziekte mechanismen te bestuderen, nieuwe medicijnen verbindingen te screenen, en om nieuwe therapieën voor motor neuron ziekten zoals spinale musculaire atrofie (SMA) en amyotrofische laterale sclerose te ontwikkelen ( ALS). Veel van de huidige protocollen maken gebruik van een combinatie van retinoïnezuur (RA) en sonic hedgehog (Shh) om de muis embryonale stamcellen (MES) cellen differentiëren tot neuronen 1-4. Echter, de differentiatie efficiëntie van MES cellen in motorische neuronen alleen een ontmoeting met matig succes. We hebben een twee-staps differentiatie protocol nr. 5 die aanzienlijk de differentiatie-efficiency verbetert in vergelijking met de huidige vastgestelde protocollen. De eerste stap is om de neuralization proces te versterken door het toevoegen van Noggin en fibroblast groeifactoren (FGF). Noggin is een bot morfogenetisch proteïne (BMP) antagonist en is betrokken bij neurale inductie eenolgens het standaard model van de neurogenese en resulteert in de vorming van de voorste neurale patronen 6. FGF-signalering werkt synergetisch met Noggin in het induceren van zenuwweefsel vorming door het bevorderen van een posterior neurale identiteit 7-9. In deze stap, werden cellen MES voorbehandeld met Noggin, bFGF, en FGF-8 twee dagen differentiatie bevorderen richting neurale lijnen. De tweede stap is het induceren van motorneuron specificatie. Noggin / FGF blootgesteld MES cellen werden geïncubeerd met RA en een Shh agonist, gladgestreken agonist (SAG), nog 5 dagen om de motor neuron generatie te vergemakkelijken. Om de differentiatie van MESs volgen in motorische neuronen, hebben we gebruik gemaakt van een ES-cellijn afgeleid van een transgene muis te drukken eGFP onder de controle van de motor neuron-specifieke promoter Hb9 1. Met behulp van deze robuuste protocol, behaalden we 51 ± 0,8% van differentiatie efficiëntie (n = 3, p &lt;0,01, Student&#39;s t-test) 5. Resultaten van immunofluorescentiekleuring toonde aan dat GFP +-cellen van de motor neuron specifieke markers, Islet-1 en choline acetyltransferase (ChAT) uit te drukken. Onze twee-staps differentiatie-protocol zorgt voor een efficiënte manier om MES-cellen differentiëren in spinale motorische neuronen.
