Mycobacterium tuberculosis, det ætiologiske agens for human tuberkulose, har en usædvanlig evne til at overleve mod miljømæssige påvirkninger, herunder antibiotika. M. tuberkulose, men fængslende kræver specifikke betingelser for at danne modne biofilm, især 09:01 forholdet headspace: medier samt begrænset udveksling af luft med atmosfæren 9. Vi har beskrevet en protokol for en svækket stamme af M. tuberkulose, mc 2 7000, med deletion i de to loci, panCD og RD1, som er kritiske for in vivo vækst af patogenet 9. Denne stamme kan sikkert bruges i en BSL-2 inddæmning for at forstå den grundlæggende biologi tuberkulose patogenet og dermed undgå kravet om et dyrt BSL-3 anlæg. Fremgangsmåden kan udvides, med passende modifikation i medier, til at vokse biofilm af andre dyrkbar mykobakteriestammer. Samlet set vil en ensartet protokol af dyrkning af mycobakterielle biofilm hjælpe efterforskerne interesseret i at studere de grundlæggende elastiske egenskaber mykobakterier.
