Herpes simplex virus type 1 (HSV-1) wordt een leven lang latente infectie in de perifere neuronen. Daarnaast dierproeven is kostbaar, tijdrovend, en beperkt in termen van beschikbare opties voor het manipuleren van gastheerprocessen. Met name lytische mRNAs, eiwitten en infectieuze virus meer detecteerbaar, zelfs in de afwezigheid van selectie, maar latentie-geassocieerde transcript (LAT) expression blijft in neuronale kernen. Virale genomen worden gehandhaafd op gemiddeld aantal kopieën van 25 per neuron en kan worden geïnduceerd om productief te repliceren door te interfereren met PI3-kinase / Akt signalisatie of de eenvoudige intrekking van zenuwgroeifactor 1.
