Vi har utviklet en cellebasert høy tetthet microarray, Ca BChip, bestående av nanoliter volumer av Candida albicans biofilm. Den microarray ble trykt på endret glass underlag, som tillot robust feste kollagen gel flekker samtidig som det gir hydrofobisitet nødvendig for en ikke-spredning, halvkuleformet 3D gel. Candida albicans fortsatt den viktigste agens av candidiasis, som for tiden representerer den fjerde vanligste nosokomiale blodbanen infeksjon i amerikanske sykehus 1. Disse opportunistiske infeksjoner utgjør en økende trussel for et økende antall av kompromitterte individer, og bære uakseptabelt høy dødelighet. Dette er delvis på grunn av den begrensede arsenal av soppdrepende medisiner, men også til utvikling av resistens mot de mest brukte soppdrepende midler. Videre kompliserer behandlingen er det faktum at et flertall av manifestasjoner av candidiasis er forbundet med dannelse av biofilm, og celler innenfor disse biofilmer viser økte nivåer av motstand til mest klinisk brukte antimykotika 2. Her beskriver vi utviklingen av en høy tetthet microarray som består av C. albicans nano-biofilm, som vi har kalt Ca BChip tre. Kort fortalt er en robot microarrayer brukt to print gjærceller av C. albicans inn på et fast underlag. Under utskrift er de gjærceller innelukket i en tredimensjonal matrise med et volum så lavt som 50 NL og immobilisert på et glass substrat med en passende belegg. Etter innledende utskriften er de lysbildene inkuberes ved 37 ° C i 24 timer for å tillate biofilm utvikling. I denne perioden flekkene vokse til fullt utviklede &quot;nano-biofilmer&quot; viser at typiske strukturelle og fenotypiske egenskaper knyttet moden C. albicans biofilm (dvs. morfologisk kompleksitet, tredimensjonale arkitektur og legemiddelresistens) 4. Totalt sett er Ca BChip sammensatt på ~ 750 likeverdige og romlig tydelig biofilm, med den ekstra fordelen at flere sjetonger kan skrives ut og behandles samtidig. Celleviabilitet anslås ved å måle fluorescerende intensiteten FUN1 metabolsk flekken med en microarray scanner. Dette sopp chip er ideelt for use i ekte high-throughput screening for soppdrepende medisiner. Sammenlignet med gjeldende standarder (dvs. 96-brønnen microtiter plate modell av biofilmdannelse 5), de viktigste fordelene av sopp biofilm chip er automatisering, miniatyrisering, besparelser i mengde og kostnad av reagenser og analyser tid, samt eliminering av arbeidskraft intensive trinn. Vi mener at en slik chip vil gi en betydelig raskere soppdrepende medisiner prosess.
