combinatorial synthesis of and high-throughput protein release from polymer film and nanoparticle libraries

Polyanhydriden zijn een klasse van biomaterialen met uitstekende biocompatibiliteit en drug delivery mogelijkheden. Terwijl ze uitvoerig bestudeerd met gebruikelijke een-sample-at-a-time synthesetechnieken, een recent high-throughput aanpak ontwikkeld waardoor de synthese en het testen van grote bibliotheken van polyanhydriden 1. Dit bevordert een efficiënte optimalisatie-en ontwerpproces van deze biomaterialen voor drugs-en toediening van vaccins toepassingen. De werkwijze in dit werk beschrijft de combinatorische synthese van biologisch afbreekbare polyanhydride film en nanodeeltjes bibliotheken en high-throughput detectie van proteïneafgifte uit deze bibliotheken. Bij deze robot bediende methode (figuur 1), zijn lineaire actuatoren en spuitpompen gecontroleerd door LabVIEW, die een hands-free geautomatiseerd protocol maakt, waardoor fout van de gebruiker. Bovendien deze werkwijze maakt de snelle fabricage van micro-schaal polymeer libraries, redvóór het eerste batchgrootte terwijl resulteert in de creatie van Multivariant polymeersystemen. De werkwijze hierin ontwikkelde combinatorische maakt high-throughput polymeersynthese en fabricage van eiwit beladen nanodeeltjes en film libraries, die op hun beurt in vitro en in vivo worden gescreend optimaliseren van prestaties biomateriaal.
