combinatorial synthesis of and high-throughput protein release from polymer film and nanoparticle libraries

Polyanhydride sind eine Klasse von Biomaterialien mit exzellenten Biokompatibilität Droge Lieferfähigkeit. Während sie wurden ausgiebig mit der studierten herkömmlichen Ein-Stichproben-at-a-time-Synthese-Technologien, eine neuere High-Throughput-Ansatz entwickelt ermöglicht die Synthese und Erprobung von großen Bibliotheken Polyanhydride 1. Dies erleichtert eine effizientere Optimierung und Design-Prozess dieser Biomaterialien für Drogen-und Impfstoff-Delivery-Anwendungen. Das Verfahren in dieser Arbeit beschreibt die kombinatorische Synthese von biologisch abbaubaren Polyanhydrid Film-und Nanopartikel-Bibliotheken und die Hochdurchsatz-Detektion von Protein-Freisetzung aus diesen Bibliotheken. In diesem Roboter betriebene Verfahren (Abbildung 1) werden lineare Aktuatoren und Spritzenpumpen von LabVIEW, die eine Freisprecheinrichtung automatisierten Protokoll ermöglicht, wodurch Anwenderfehler gesteuert. Darüber hinaus ermöglicht dieses Verfahren die rasche Herstellung von Mikro-Maßstab Polymer Bibliotheken, rotenucing die Chargengröße während was zur Schaffung multivariante Polymersystemen. Diese kombinatorische Ansatz zur Synthese von Polymeren ermöglicht die Synthese von bis zu 15 verschiedene Polymere in eine äquivalente Menge an Zeit, die für ein Polymer konventionell synthetisieren würde. Darüber hinaus kann die kombinatorische Bibliothek in freilassen Polymer oder Protein-beladenen Geometrien einschließlich Filmen oder Nanopartikel beim Auflösen des Polymers Bibliothek in einem Lösungsmittel und Ausfällen in einem Nichtlöser (z. Nanopartikel) oder durch Vakuumtrocknung (für Folien) hergestellt werden. Nach dem Laden eines Fluorochrom-konjugierten Proteins in die Polymer-Bibliotheken kann Protein Freisetzungskinetik bei hohem Durchsatz untersucht werden mit Hilfe eines Fluoreszenz-basierten Detektionsverfahren (Abbildungen 2 und 3), wie zuvor beschrieben ein. Diese kombinatorische Plattform wurde mit konventionellen Methoden validiert wurden 2 und das Polyanhydrid Film-und Nanopartikel-Bibliotheken wurden mit geprägt <sup&gt; 1 H-NMR-und FTIR. Die Bibliotheken haben für Protein Freisetzungskinetik, Stabilität und Antigenität durchsucht, in vitro zelluläre Toxizität, Zytokin-Produktion, Oberflächenmarker Ausdruck, Adhäsion, Proliferation und Differenzierung, und in vivo Bioverteilung und Mucoadhäsion 1-11. Die kombinatorischen Methode entwickelt hierin ermöglicht Hochdurchsatz-Polymer-Synthese und Herstellung von Protein-beladenen Nanopartikel und Filmbibliotheken, welche wiederum können, in vitro und in vivo zur Optimierung der Leistung Biomaterial gescreent werden.
