Biologisch abbaubare Nanopartikel als vielseitige Plattform für die Konzeption und Umsetzung von neuen intranasale Impfstoffe gegen respiratorische Infektionskrankheiten aufgetaucht. Insbesondere Nanopartikel Polyanhydrid aus der aliphatischen Sebacinsäure (SA), das aromatische 1,6-Bis (p-carboxyphenoxy) hexan (BER), oder die amphiphilen 1,8-Bis (p-carboxyphenoxy) -3,6-dioxaoctan (CPTEG) haben einzigartige Masse und Oberflächenerosion Kinetik 1,2 und kann genutzt werden, um langsam freisetzen funktionelle Biomoleküle (zB Protein-Antigene, Immunglobuline, usw.) sind in vivo 3,4,5 werden. Diese Nanopartikel besitzen auch intrinsische Adjuvans-Aktivität, so dass sie eine ausgezeichnete Wahl für einen Impfstoff-Delivery-Plattform 6,7,8. Um die Mechanismen für die Aktivierung der angeborenen Immunität nach intranasaler Schleimhaut Impfung aufzuklären, muss man beurteilen, die molekularen und zellulären Reaktionen des Antigens pübelnehmend Zellen (APC) verantwortlich für die Initiierung Immunantworten. Dendritische Zellen sind die wichtigsten APCs in leitenden Atemwege vor, während Alveolarmakrophagen (AMɸ) im Lungenparenchym 9,10,11 überwiegen. AMɸ sind hoch effizient bei der Räumung in die Lunge von mikrobiellen Krankheitserregern und Zelltrümmer 12,13. Darüber hinaus spielt dieser Zelltyp eine wertvolle Rolle bei der Beförderung von mikrobiellen Antigenen zu den Lymphknoten, die ein wichtiger erster Schritt bei der Initiation einer adaptiven Immunantwort 9 ist. AMɸ auch zum Ausdruck bringen erhöhte Werte von angeborenen Mustererkennung und Scavenger-Rezeptoren, sezernieren pro-inflammatorischen Mediatoren und prime naiven T-Zellen 12,14. Ein relativ reine Population von AMɸ (z. B. größer als 80%) kann ebenfalls in Lungenspülung für Studie im Labor erhalten werden. Wohnhaft AMɸ von immunkompetenten Tieren geerntet werden einen repräsentativen Phänotyp der Makrophagen, wird encounter die Partikel-basierten Impfstoff in vivo. Hier beschreiben wir die Protokolle verwendet werden, um Ernte und Kultur AMɸ von Mäusen zu untersuchen und die Aktivierung der Makrophagen-Phänotyp nach Behandlung mit Polyanhydrid Nanopartikel in vitro.
