Tumor heterogeniteit is een fundamenteel kenmerk ondersteunende tumor robuustheid en presenteert een centrale belemmering voor de ontwikkeling van therapeutische strategieën 1. Te overwinnen het probleem van tumor heterogeniteit is het noodzakelijk dat assays en instrumenten waarmee fenotypische, (epi) genetische en functionele identificatie en karakterisatie van tumor subpopulaties die specifieke ziekte pathologieën rijden en klinisch relevante doelen. Het is nu bekend dat tumoren vertonen verschillende deelfracties van cellen met verschillende frequenties van celdeling, en de functionele criterium dat langzaam fietsen positief geassocieerd met tumorvorming vermogen in verschillende kankers waaronder die van de hersenen, borst, huid en pancreas en leukemie 2-8. De fluorescente kleurstof carboxyfluoresceïne succinimidylester (CFSE) is gebruikt voor het volgen van de verdeling frequentie van cellen in vitro en in vivo in bloed overgedragen tumors en vaste tumoren zoals 2,7,8 glioblastoma. De cel-permeant niet fluorescerend pro-drug van CFSE omgezet door intracellulaire esterasen in een fluorescente verbinding, die binnen cellen vastgehouden door covalente binding aan eiwitten door reactie van de succinimidyl groep met intracellulaire aminegroepen vormen stabiele amidebindingen 9. De fluorescerende kleurstof wordt gelijkelijk verdeeld tussen dochtercellen op divisies leidt tot de halvering van de fluorescentie-intensiteit bij elke celdeling. Dit maakt het volgen van celcyclus frequentie tot acht tot tien ronden van sector 10. CFSE retentie capaciteit werd gebruikt met hersentumor cellen te identificeren en te isoleren van een langzaam fietsen subpopulatie (top 5% kleurstof-behoud van cellen) aangetoond worden verrijkt in kankerstamcel activiteit 2. Dit protocol beschrijft de techniek van gekleurde cellen met CFSE en isolatie van afzonderlijke populaties in een cultuur van menselijke glioblastoom (GBM)-afgeleide cellen bezitten verschillende divisie tarieven met behulp van flowcytometrie 2. De techniek heeft gediend te identificeren en een hersentumor langzaam cycling populatie van cellen te isoleren door hun vermogen om de CFSE labeling behouden.
