De vooruitgang in microfabricage technieken hebben het mogelijk de productie van goedkope en reproduceerbare microfluïdische systemen voor het uitvoeren van biologische en biochemische experimenten op micro-en nanoschaal 1,2. Daarnaast zijn microfluidics ook specifiek gebruikt om kwantitatief te analyseren hematologische en microvasculaire processen, vanwege hun vermogen om eenvoudig controle de dynamische vloeibare milieu en biologische omstandigheden 3-6. Als zodanig hebben de onderzoekers meer recent gebruikt microfluïdische systemen bloedcellen vervormbaarheid, bloedcel aggregatie, microvasculaire doorbloeding, en bloed cel-endotheliale cel interacties 6-13 te bestuderen. Echter, deze microfluïdische systemen ofwel geen rekening gehouden met gekweekte endotheelcellen of waren groter dan sizescale relevante microvasculaire pathologische processen. Een microfluïdische platform met gekweekte endotheelcellen die nauwkeurig recapituleert de cellulaire, fysieke en hemodynAmic omgeving van de microcirculatie is nodig om ons begrip van de onderliggende biofysische pathofysiologie van hematologische ziekten die de microvasculatuur te betrekken bevorderen. Hier melden wij een methode om een &quot;endothelialized&quot; te creëren in vitro model van de microvasculatuur, met behulp van een eenvoudige, enkel masker microfabricage proces in combinatie met standaard endotheelcellen cultuur technieken, om pathologische biofysische microvasculaire interacties die plaatsvinden in de hematologische ziekte te bestuderen. Deze &quot;microvasculatuur-on-a-chip&quot; biedt de onderzoeker met een robuuste test die goed bestuurt zowel biologische als biofysische voorwaarden en wordt bediend met een standaard spuit pomp en helderveld / fluorescentie microscopie. Parameters zoals microcirculatie hemodynamische omstandigheden, endotheelcellen type bloedcel type (n) en de concentratie (s), drugs / remmende concentratie etc. kunnen allemaal eenvoudig te bedienen. Als zodanig is onze microsysteem biedteen methode om kwantitatief te onderzoeken ziekte processen waarin microvasculaire doorstroming wordt belemmerd als gevolg van veranderingen in celadhesie, aggregatie, en vervormbaarheid, een vermogen beschikbaar met bestaande tests.
