TIRF microscopie is de techniek bij uitstek om beeld perifere eiwitten. De lage penetratie diepte van de evanescente veld minimaliseert van onscherpe licht, wat leidt tot een zeer goede signaal-ruisverhouding en maakt het mogelijk data-acquisitie met een hoge frame rates, of beeldvorming van heel zwak tot expressie gebrachte eiwitten. TIRF microscopie heeft zich ontpopt als een krachtig imaging technologie om spatio-temporele dynamiek van fluorescerende moleculen in vitro te bestuderen en in levende cellen 1. De optische verschijnsel van totale interne reflectie optreedt wanneer licht van een medium met hoge brekingsindex in een medium met lage brekingsindex een hoek groter dan een kenmerk grenshoek (bijvoorbeeld dichter bij parallel zijn met de grens). Hoewel al het licht wordt gereflecteerd terug onder zulke omstandigheden, is een vergankelijk golf gecreëerd die zich voortplant over en langs de grens, die exponentieel vervalt met de afstand, en alleen doordringt monster gebieden die 100-200 nm de buurt van de interface 2. Naast het leveren van een superieure axiale resolutie, de verminderde excitatie van onscherpe fluoroforen zorgt voor een zeer hoge signaal-ruis-ratio&#39;s en minimaliseert schadelijke effecten van fotobleken 2,3. Omdat het een groothoek techniek, TIRF maakt het ook mogelijk een snellere beeldverwerking acquisition dan de meeste op basis van het scannen van confocale setups. Op het eerste gezicht, de lage indringdiepte van TIRF niet compatibel is met beeldvorming van bacteriële en fungale cellen, die vaak zijn omgeven door een dikke celwanden. Integendeel, hebben we ontdekt dat de celwanden van gist en bacteriële cellen eigenlijk de bruikbaarheid van TIRF verbeteren en vergroten het bereik van waarneembare structuren 4-8. Veel cellulaire processen kan dus direct worden benaderd door TIRF in kleine, eencellige micro-organismen, die vaak bieden krachtige genetische manipulatie technieken. Dit stelt ons in staat om te presteren in vivo experimenten biochemie, waar kinetiek van eiwit interacties en activiteiten die direct kan worden beoordeeld in levende cellen. We beschrijven hier de individuele stappen die nodig zijn om hoge kwaliteit TIRF beelden te verkrijgen voor Saccharomyces cerevisiae of Bacillus subtilis cellen. Wij wijzen erop verschillende problemen die kunnen Affect TIRF visualisatie van fluorescerende probes in cellen en illustreren de procedure met een aantal toepassingsvoorbeelden. Tot slot laten we zien hoe TIRF beelden kunnen verder worden verbeterd met behulp van gevestigde beeld restauratietechnieken.
