Retrograde Markierung von Neuronen ist ein Standard anatomische Methode, die auch 1,2 verwendet wurde, um Calcium und spannungsabhängigen Farbstoffen in Neuronen 3-6 geladen. Im Allgemeinen werden die Farbstoffe als feste Kristalle oder durch lokale Injektion mit Druck Glaspipetten aufgebracht. Dies kann jedoch zu einer Verdünnung des Farbstoffs und vermindertem Markierungsintensität führen, insbesondere wenn mehrere Stunden Farbstoffdiffusion erforderlich sind. Hier zeigen wir eine einfache und kostengünstige Technik zur Einführung fluoreszierenden und Ionen-sensitiven Farbstoffen in Neuronen ein Polyethylen Saugpipette mit dem Farbstoff-Lösung gefüllt. Dieses Verfahren bietet eine zuverlässige Möglichkeit zur Aufrechterhaltung einer hohen Konzentration des Farbstoffs in Kontakt mit Axone in der Ladevorgang.
