Derzeit Methoden der super-hochauflösenden Mikroskopie, zB auf strukturierte Beleuchtung 7 oder auf Einzelmoleküldetektion 8-10 sowie stimulierte Emission Depletion (STED) Mikroskopie 11 sind von großem Interesse basiert. Keine dieser Techniken erlaubt jedoch eine axiale Auflösung unterhalb von 50 nm aufweist. Darüber hinaus relativ hohe Bestrahlungsstärke von 50100 W / cm 2...

Wenn ein Lichtstrahl durch ein Medium ausbreitet der Brechungsindex n 1 eine Schnittstelle erfüllt, um ein zweites Medium mit einem Brechungsindex n 2 &lt;n 1 tritt Totalreflexion an alle Einfallswinkel Θ, der größer als der kritische Winkel Θ c = sind arcsin (n 2 / n 1). Gleichung 3 enthalten kann analytisch gelöst werden, wenn die Konzentration c (z) von Fluorophoren vorliegt, gilt als nahezu konstant - Entweder überhaupt Koordinaten z ≥ Δ, im Zytoplasma von Zellen mit einem Abstand Δ von der Schnittstelle zB. Für die Membran Marker Laurdan in diesem Papier verwendet (s. unten) die Auswertung nach Gl. 5 durchgeführt wurde. Wie bereits berichtet 3, Bildaufnahme und Auswertung benötigt - Homogene Verteilung von Anregungslicht über die Probe, - Gültigkeit einer 2-Schichten-Modells (mit den Brechungsindizes n 1 und n 2 für das Substrat und das Zytoplasma, jeweils). Dies gilt, wenn die Plasmamembran ist sehr dünn, und wenn die Schicht aus dem extrazellulären Medium (zwischen der Zelle und dem Substrat) klein ist im Vergleich zur Wellenlänge des einfallenden Lichts. Zusätzlich kann eine moderate numerische Apertur(Typischerweise A ≤ 0,90) des Mikroskops Objektivlinse ist vorteilhaft, Abweichungen der Helligkeit aufgrund Anisotropie von Strahlung im nahen Feld des Dielektrikum-Grenzfläche zu vermeiden.

Oberflächentopologie, z. B. von Zellen wachsen auf einem Substrat, mit Nanometer-Präzision durch unterschiedlichen Winkeln Total Internal Reflection-Fluoreszenzmikroskopie (VA-TIRFM) bestimmt. Zellen werden auf transparente Folien kultiviert, gewaschen und mit einem fluoreszierenden Marker homogen in ihrem Plasma Membran verteilt. Beleuchtungssystem tritt durch einen parallelen Laserstrahl unter variablen Winkeln der inneren Totalreflexion (TIR) ​​mit unterschiedlichen Eindringtiefen der evaneszenten elektromagnetischen Feldes. Aufzeichnung von Bildern bei Bestrahlung Fluoreszenz bei etwa 10 unterschiedlichen Winkeln ermöglicht, Zell-Substrat-Abstände mit einer Genauigkeit von wenigen Nanometern berechnen. Unterschiede der Haftung zwischen verschiedenen Zelllinien, zB Krebszellen und weniger malignen Zellen werden somit bestimmt. Darüber hinaus können eventuelle Änderungen der Zelladhäsion auf chemische oder photodynamischen Behandlung geprüft. Im Vergleich mit anderen Methoden der super-hochauflösenden Mikroskopie Belichtung gehalten wirdsehr klein, und keine Beschädigung der lebenden Zellen erwartet wird.
