We toonden aan dat volwassen subependymale NPCs snel en gericht kathodische migratie ondergaan in vitro bij blootstelling aan een extern aangebracht dcEF. In dit protocol beschrijven we ons lab de technieken voor het creëren van een eenvoudige en effectieve galvanotaxis test voor hoge resolutie lange termijn observatie van gericht cellichaam translocatie (migratie) op een single-cell niveau. Deze test zou geschikt zijn voor het onderzoeken van de mechanismen die in cellulaire transductie dcEF motiliteit reguleren door het gebruik van transgene of knock-out muizen, korte interfererende RNA, of specifieke receptor agonisten / antagonisten.

Kinematische analyse blijkt dat in de aanwezigheid van een 250 mV / mm dcEF, ongedifferentieerde NPCs zeer gericht en snelle galvanotaxis naar de kathode (Figuur 5A, Movie 1) vertonen. In afwezigheid van een dcEF wordt willekeurige beweging van de cellen waargenomen (Figuur 5B, Movie 2). Dit veldsterkte&gt; 98% van ongedifferentieerde NPCs migreren voor de gehele 6-8 hr waarvoor zij worden afgebeeld, en aangezien er dode cellen migreren niet suggereert dat zij levensvatbaar blijven gedu...
