Specifieke leden van de regeling actieve gelijkrichter kalium (Kir) kanaal familie worden gepostuleerd drug targets voor een verscheidenheid van aandoeningen, waaronder hypertensie, atriumfibrilleren, en pijn 1,2. Voor het grootste deel, echter, is de vooruitgang in de richting van het begrijpen van hun therapeutisch potentieel of zelfs elementaire fysiologische functies is vertraagd door het gebrek aan goede farmacologische tools. De Molecular Libraries Probes Production Center Network (MLPCN), ondersteund door de National Institutes of Health (NIH) Gemeenschappelijk Fonds heeft mogelijkheden gecreëerd voor academische wetenschappers sonde ontdekking campagnes te starten voor moleculaire targets en signaalwegen die behoefte hebben aan een betere farmacologie 6. De MLPCN biedt onderzoekers toegang tot de industrie-schaal screening centra en medicinale chemie en informatica ondersteuning op laag-moleculaire sondes om de functie van genen en genproducten netwerken toe te lichten te ontwikkelen. De kritische stap in het verkrijgen van toegang tot de MLPCN is de ontwikkeling van een robuuste target-of route-specifieke assay die vatbaar voor high-throughput screening (HTS). Hier beschrijven we hoe u een fluorescentie gebaseerde thallium (Tl +) flux ASSA ontwikkeleny van Kir kanaalfunctie voor high-throughput screening verbinding 7,8,9,10. De assay is gebaseerd op de permeabiliteit van de K + kanaalporie de K + Tl + congener. Een commercieel verkrijgbare fluorescerende Tl + reporter kleurstof wordt gebruikt flux van Tl + detecteren door de porie. Er zijn minstens drie beschikbare kleurstoffen die geschikt zijn voor Tl + flux assays: BTC, FluoZin-2 en FluxOR 7,8. Dit protocol beschrijft assay-ontwikkeling met behulp FluoZin-2. Hoewel oorspronkelijk ontwikkeld en verkocht als een zink indicator, FluoZin-2 vertoont een robuuste en dosisafhankelijke toename in fluorescentie-emissie bij Tl + binding. We begonnen te werken met FluoZin-2 voor FluxOR beschikbaar was 7,8 en zijn doorgegaan met dat 9,10 doen. De stappen in assay ontwikkeling zijn in wezen identiek voor alle drie kleurstoffen en gebruikers moeten bepalen welke kleurstof meest geschikt is voor de specifieke needs. We bespreken ook de test de prestaties van benchmarks die moeten worden bereikt om in aanmerking komen voor de toegang tot het MLPCN. Sinds Tl + gemakkelijk doordringt de meeste K + kanalen, moet de test kunnen worden aangepast aan de meeste K + kanaal doelstellingen.

Het gebruik van een tetracycline-induceerbaar expressiesysteem een handige interne controle voor het onderscheid Tl + flux via endogene routes en Kir kanaal plaats.
