Xenopus laevis neurale explantaten sturen neurieten in een robuuste wijze door 24 uur na het uitplaten van de laminine / poly-lysine substraat als de omstandigheden nodig. Met dit substraat groei kegels zijn zeer beweeglijke en kunnen axon lengtes bereiken 1 mm, die in alle richtingen naar buiten uit het explantaat, hoewel typische lengtes 100 urn of meer. Als neurieten niet groeien, zijn er een beperkt aantal redenen het geval. Een mogelijkheid is dat de neurale explantaten niet goed hechten aan de schotel. Zo...

Het complexe axon begeleiding wordt voornamelijk veroorzaakt door de groei cone, dat de dynamische beweeglijke structuur aan het uiteinde van de groeiende axon. Tijdens axon uitgroei, moet de groei kegel integreren meerdere bronnen van begeleiding cue informatie voor het moduleren van de cytoskelet om de groei kegel vooruit te stuwen en nauwkeurig navigeren naar de specifieke doelstellingen ervan vinden 1. Hoe deze integratie vindt plaats op het cytoskelet niveau is nog steeds in opkomst, en onderzoek van het cytoskelet eiwitten en effector dynamiek binnen de groei conus kan de opheldering van deze mechanismen toe te staan. Xenopus laevis groei kegels zijn groot genoeg (10-30 micron in diameter) aan op hoge resolutie beeldvorming van levende cytoskelet dynamiek (bijvoorbeeld 2-4) en zijn gemakkelijk te isoleren en in een laboratoriumomgeving manipuleren vergelijking met andere vertebraten. De kikker is een klassiek model voor de ontwikkelingsbiologie neurobiologie studies en belangrijke vroege inzichten in de groei kegel microtubule dynamiek werden in eerste instantie gevonden met behulp van dit systeem 5-7. In deze werkwijze 8 worden verzameld en eitjes bevrucht in vitro, geïnjecteerd met RNA dat codeert fluorescent gelabeld cytoskelet fusie-eiwitten of andere constructies om genexpressie te manipuleren, en vervolgens liet men ontwikkelen om de neurale buis fase. Neurale buizen worden geïsoleerd door dissectie en vervolgens gekweekt en groei kegels op uitgroeiende neurieten worden afgebeeld. In dit artikel beschrijven we hoe u deze methode, waarvan het doel is om de cultuur Xenopus laevis groei kegels voor de volgende afbeelding in hoge resolutie analyse uit te voeren. Terwijl wij het voorbeeld van + TIP fusieproteïne EB1-GFP, kan deze werkwijze worden toegepast op een aantal eiwitten om hun gedrag verduidelijken in de groeikegel.
