De wijziging van virusdeeltjes heeft een aanzienlijk deel van de aandacht voor zijn enorme potentieel voor invloed gentherapie, oncolytische toepassingen en de ontwikkeling van vaccins. 1,2,3 huidige aanpak op de wijziging van virale oppervlakken, die meestal op basis van de genetica, vaak last van demping van het virus productie, infectiviteit en cellulaire transductie. 4,5 Met behulp van chemoselectieve klik chemie, hebben we een duidelijke alternatieve benadering, waarin deze aspecten omzeilt, terwijl de resterende zowel zeer flexibel en toegankelijk. 1,2 Het doel van dit protocol is om aan te tonen de effectiviteit van het gebruik van bioorthogonal klik chemie aan de oppervlakte van adenovirus type 5 deeltjes te wijzigen. Deze twee stappen kunnen zowel therapeutisch 1 of analytisch, 2,6 worden gebruikt als het zorgt voor chemoselectieve ligatie van targeting moleculen, kleurstoffen of andere moleculen van belang op eiwittengelabelde met azide tags. De drie grote voordelen van deze methode zijn dat (1) metabole etikettering weinig tot geen invloed op de virale fitness, 1,7 laat zien (2) een brede waaier van effector liganden kan worden gebruikt, en (3) het is opmerkelijk snel, betrouwbaar en makkelijk te bereiken. 1,2,7 In de eerste stap van deze procedure worden adenoviruspartikels geproduceerd met een azidohomoalanine (Ha een methionine surrogaat) of onnatuurlijke suiker O-gekoppelde N-azidoacetylglucosamine (O-GlcNAz), die beide de azide (N-3) functionele groep. Na zuivering van azide gemodificeerde virus deeltjes wordt een alkyn probe met de fluorescerende TAMRA groep geligeerd in chemoselectieve wijze de pre-gemerkte eiwitten of glycoproteïnen. Tenslotte is een SDS-PAGE analyse uitgevoerd om de succesvolle ligatie van de probe aangetoond op het virale capside eiwitten. Aha opname wordt getoond aan alle virale capside-labeleiwitten (Hexon, Penton en Fiber), terwijl O-GlcNAz opname resulteert in de etikettering van Fiber alleen. In deze ontwikkelende gebied zijn er verschillende methoden voor het azide-alkyn ligatie met succes ontwikkeld, maar alleen de twee die we hebben gevonden zijn meest handige handleiding worden aangetoond - stam-bevorderd azide-alkyn cycloadditie (SPAAC) en koper-gekatalyseerde azide-alkyn cycloadditie (CuAAC) onder zuurstofarme atmosfeer.
