Autoimmune fenomener oppstår hos friske individer, men når selv toleranse mislykkes, kan autoimmun reaksjon resultere i spesifikk patologi. Ifølge Witebsky største postulater, ett av kriteriene i å diagnostisere en sykdom som autoimmun er gjengivelse av sykdommen i forsøksdyr ved passiv overføring av autoantistoffer. For Epidermolysis bullosa acquisita (EBA), en prototypic organspesifikk autoimmun sykdom av hud og slimhinner, ble flere eksperimentelle modeller nylig etablert. I dyremodellen beskrevet i vår foreliggende arbeid, renset IgG-antistoffer mot en strekning av 200 aminosyrer (aa 757-967) av kollagen VII er injisert gjentatte ganger i mus reproduserende blistering fenotypen samt histo-og immunopathological funksjoner karakteristiske til menneske EBA 1. Fullverdig utbredt sykdom ses vanligvis 5-6 dager etter den første injeksjonen og omfanget av sykdommen korrelerer med dosen av den administrerte collagen VII-spesifikk IgG. Vevsskaden (blæredannelse) i den eksperimentelle EBA avhengig rekruttering og aktivering av granulocytter ved vev-bundet autoantistoffer 2, -4. Derfor gir denne modell for disseksjon av granulocytt-avhengige inflammatoriske pathway involvert i autoimmune vevsskade, som modell reproduserer bare T-celle-uavhengig fase av efferent autoimmun respons. Videre er dens verdi understreket av en rekke studier som viser blemmeaktig induserende potensialet av autoantistoffer in vivo og undersøke mekanismen av blæredannelse i 1,3 EBA, -6. Til slutt vil denne modellen i stor grad forenkle utviklingen av nye anti-inflammatorisk behandling i autoantistoff-induserte sykdommer. Totalt sett er passiv overføring dyremodell av EBA et tilgjengelig og lærerikt sykdom modell og vil hjelpe forskerne å analysere ikke bare EBA patogenese, men å svare grunnleggende biologisk og kliniskessensielle autoimmunitet spørsmål.
