A morte celular programada (PCD) ocorre em adultos para manter a homeostase do tecido normal e durante o desenvolvimento embrionário de moldar os tecidos e órgãos 1,2,6,7. Durante o desenvolvimento, produtos químicos tóxicos ou alterações genéticas podem causar um aumento na PCD ou mudar os padrões de PCD, resultando em anomalias do desenvolvimento e defeitos de nascimento 3-5. Para compreender a etiologia de tais defeitos, o estudo de embriões podem ser complementados com os ensaios in vitro que utilizam diferenciação estaminais embrionárias (ES) células. A apoptose é uma forma bem estudado de PCD que envolve tanto intrínsecos e extrínsecos de sinalização para ativar a enzima caspase cascata. Alterações celulares características incluem blebbing membrana, encolhimento nuclear, e fragmentação de DNA. Outras formas de PCD não envolvem a activação das caspases e pode ser o resultado final de autofagia prolongada. Independentemente da via de PCD, as células que morrem precisam de ser removidos. Em adultos, as células do sistema imunológico performe esta função, enquanto que em embriões, em que o sistema imune não desenvolveu ainda, a remoção ocorre por um mecanismo alternativo. Este mecanismo envolve as células vizinhas (chamados &quot;não profissionais fagócitos&quot;) assumindo um papel-fagocítica eles reconhecem o &#39;me comer&#39; sinal na superfície da célula a morrer e que engolem 8-10. Em contraste com o azul de anilina, que difunde e é dissolvido por solventes, Lyso Tracker Red DND-99 é corrigível, brilhante e estável. Aqui descrevemos protocolos utilizando o corante de olhar para PCD no normal e Sonic Hedgehog embriões de camundongos nulos. Além disso, demonstramos análise de PCD em diferenciar as culturas de células ES e apresentar um método de quantificação simples. Em resumo, coloração Lyso Tracker pode ser um ótimo complemento para outros métodos de detecção de PCD.
