Bij het visualiseren van de cellen met fluorescerende eiwitten zoals GFP, is het belangrijk om niet de bereiding overbelichte te veel licht. Dit kan resulteren in schade foto. We maken gebruik van 100W HBO korte boog kwik lampen voor verlichting, en we hebben ook ND (neutrale dichtheid) het gebruik van 0,8 (Chroma ND filters 0,3 en 0,5). Om te kunnen beoordelen van de kwaliteit van de reiniging goed zicht is cruciaal. Derhalve kan ND filters worden verwijderd voor een korte periode van ongeveer 20 seconden voor meerdere...

Korte generatie tijden en gemakkelijke genetische technieken maken de fruitvlieg Drosophila melanogaster een uitstekend genetisch model in fundamenteel neurowetenschappelijk onderzoek. Ion-kanalen vormen de basis van alle gedrag, omdat zij bemiddelen neuronale prikkelbaarheid. De eerste spanningsgevoelige gekloonde ionkanalen was de Drosophila spanningsgevoelige kalium kanaal Shaker 1,2. Naar het begrijpen van de rol van ionenkanalen en membraan prikkelbaarheid voor functioneren van het zenuwstelsel is het nuttig om krachtige genetische tools beschikbaar te combineren in Drosophila met in situ patch clamp opnames. Jarenlang dergelijke opnamen werden gehinderd door de kleine omvang van de Drosophila CNS. Bovendien is een robuuste omhulling van glia en collageen vormden hindernissen voor patch pipet toegang tot centrale neuronen. Het verwijderen van deze mantel is een noodzakelijke voorwaarde voor patch clamp opnamen van elke neuron in de volwassen Drosophila CNS. In de afgelopen jarenwetenschappers in staat geweest om in situ patch clamp opnames te voeren van neuronen in de volwassen hersenen 3,4 en ventrale zenuw koord van embryonale 5,6, larvale 7,8,9,10, en volwassen Drosophila 11,12,13,14. Een stabiele giga-seal is de belangrijkste voorwaarde voor een goede patch en afhankelijk van schoon contact van de patch pipet met de celmembraan lekstromen te voorkomen. Daarom moet voor gehele cellen in situ patch clamp opnames van volwassen Drosophila neuronen grondig gereinigd. In de eerste stap, het ganglionische mantel moet enzymatisch behandeld en mechanisch verwijderd om de doelcellen bereiken. In de tweede stap het celmembraan zodanig worden gepolijst dat geen laag van glia, collageen of ander materiaal kan giga-seal vorming storen. In dit artikel wordt beschreven hoe u een geïdentificeerde centrale neuron te bereiden in de Drosophila ventrale zenuw koord, de vlucht motoneuron 5 (MN5 15), voor somatische hele cell patch clamp opnames. Identificatie en zichtbaarheid van het neuron wordt bereikt door gerichte expressie van GFP in MN5. We hebben niet tot doel om de patch-clamp techniek zelf uit te leggen.
