Enkele modellen die nu bestaan ​​om realistisch te simuleren van de complexe menselijke darm micro-omgeving, waar een verscheidenheid aan interacties plaatsvinden. Deze interacties zijn tot dusver zeer moeilijk te reproduceren met in vitro assaysbehulp van gekweekte cellijnen of perifere mononucleaire bloedcellen. Bovendien muismodellen aanzienlijk verschillen in bestanddelen van het darmslijmvlies (slijmlaag organisatie commensaalbacteriën gemeenschap) ten opzichte van de menselijke darm. Aldus hebben studies van verschillende behandelingen te brengen in de klinieken voor belangrijke stress of pathologische aandoeningen zoals prikkelbare darmsyndroom, inflammatoire darmziekte of colorectale kanker moeilijk uitvoerbaar zijn. Om deze problemen aan te pakken, hebben we een nieuw systeem dat ons in staat stelt om explantaten van het menselijk darmslijmvlies dat hun in situ conditionering behouden door de gastheer microbiota en de immuunrespons te stimuleren, in een gepolariseerde fashion ontwikkeld. Gepolariseerde apicale stimulatie is van groot belang voor het resultaat van de opgewekte immuunreactie. Het is herhaaldelijk aangetoond dat dezelfde stimuli produceren totaal verschillende reacties wanneer en passeren apicale oppervlak van de intestinale epithelium, het stimuleren van epitheelcellen basolateraal of die in direct contact met de lamina propria onderdelen, schakelen het fenotype van tolerogene tot immunogeen en het veroorzaken van onnodige en overmatige ontsteking in het gebied. We bereikten gepolariseerd stimulatie door lijmen een grot cilinder die afgebakend het gebied van stimulatie op de apicale gezicht van het slijmvlies zoals zal in het protocol worden beschreven. We gebruikten dit model te onderzoeken, onder meer differentiële effecten van drie verschillende Lactobacilli stammen. We tonen aan dat dit model systeem is zeer krachtig om de immunomodulerende eigenschappen van probiotica te beoordelen bij gezonde en aandoeningen.

We erin geslaagd in het behoud van gezonde en IBD menselijk darmslijmvlies in de cultuur gedurende minstens 24 uur behouden het welzijn van het weefsel. In overeenstemming met eerdere waarnemingen 1 was dit alleen mogelijk als de meerderheid van de kweekperiode (ten minste 85% van het totaal) vond plaats in O 2, zoals het weefsel niet overleven 24 uur in gebruikelijke incubatoren (figuur 2).
