analysis of snare-mediated membrane fusion using an enzymatic cell fusion assay

Samspelet mellan SNARE (s oluble N-etylmaleimid-känslig faktor en ttachment protein re Ceptor) proteiner på vesiklar (v-Snares) och mål membran (t-Snares) katalyserar intracellulär vesikelfusion 1-4. Rekonstituering analyser är väsentliga för dissekera mekanismen och regleringen av SNARE-medierad membran fusion 5. I en cellfusion analys 6,7, är SNARE-proteiner uttrycks ektopiskt vid cellytan. Dessa &quot;vänt&quot; SNARE-proteiner drive cell-cellfusion, vilket demonstrerar att Snares är tillräckliga för att smälta cellmembran. Eftersom cellfusionen analys är baserad på mikroskopisk analys, är det mindre effektivt när det används för att analysera flera v-och t-SNARE interaktioner kvantitativt. Här beskriver vi en ny analys 8 som kvantifierar SNARE-medierade händelser cellfusion genom aktiverad expression av β-galaktosidas. Tvåkomponenter Tet-Off genuttryck systemet 9 används som ett avläsningssystem: den tetracyklin-kontrollerad transaktivator (tTA) och en reporter-plasmid som kodar för LacZ-genen under kontroll av den tetracyklin-svarselementet (TRE-LacZ). Vi transfektera tTA in i COS-7-celler som uttrycker vänt v-SNARE proteiner vid cellytan (v-celler) och transfektera TRE-LacZ in i COS-7-celler som uttrycker vänt t-SNARE proteiner vid cellytan (t-celler) . SNARE-beroende fusion av V-och T-celler resulterar i bindning av tTA till TRE, den transkriptionella aktiveringen av LacZ och expression av β-galaktosidas. Aktiviteten av β-galaktosidas kvantifieras med användning av en kolorimetrisk metod genom absorbans vid 420 nm. De vesikel-associerade membranproteiner (Vamps) är V-snaror som finns i olika post-Golgi vesikulära fack 10-15. Genom att uttrycka vamps 1, 3, 4, 5, 7 och 8 på samma nivå, jämför vi deras membranfusionaktiviteter med hjälp av enzymatisk analys cellfusion. Baserat på spektrometrisk mätning, erbjuder denna analys en kvantitativ metod för att analysera SNARE-medierad membran fusion och för hög kapacitet studier.

Att utveckla en kvantitativ analys cellfusion, tar vi fördel av den starka transkriptionsaktivering genom bindning av tTA till TRE. I frånvaro av tTA, är transkription av LacZ-genen i TRE-LacZ tyst. När tTA är närvarande, binder den till TRE och aktiverar transkriptionen av LacZ.
