De ontdekking van RNAi pathway in eukaryoten en de daaropvolgende ontwikkeling van RNAi middelen, zoals siRNA en shRNA, hebben een krachtige werkwijze voor silencing specifieke genen 1-8 voor functional genomics and therapeutics. Een belangrijke uitdaging die betrokken zijn bij RNAi gebaseerde studies is de levering van RNAi agenten gerichte cellen. Traditionele niet-virale aflevering technieken, zoals elektroporatie bulk en chemische transfectie methoden vaak niet de vereiste ruimtelijke controle productietijd en 9-12 veroorloven slechte transfectie-efficiëntie. Recente vooruitgang in chemische transfectiewerkwijzen zoals kationische lipiden, kationische polymeren en nanodeeltjes hebben geresulteerd in sterk verbeterde transfectie-efficiëntie 13. Onze lab eerder het gebruik van micro-elektrode arrays (MEA) voor plaatsspecifieke transfectie in zoogdiercellen hechtende celkweken 17-19 aangetoond. In de MEA aanpak, wordt de levering van genetische lading bereikt via lokale micro-schaal electroporatiop cellen. Een toepassing van elektrische puls om geselecteerde elektroden genereert lokaal elektrisch veld dat leidt tot elektroporatie van cellen in de regio van de gestimuleerde elektrodes. De onafhankelijke controle van de micro-elektroden biedt ruimtelijke en temporele controle transfectie en kunnen meerdere transfectie gebaseerde experimenten worden uitgevoerd op dezelfde cultuur verhogen van de doorvoer en experimentele verminderen cultuur naar cultuur variabiliteit. Hier beschrijven we de experimentele opstelling en het protocol voor gerichte transfectie van het aanklevende HeLa cellen met een fluorescent gelabeld gecodeerde volgorde siRNA met behulp van elektroporatie. Hetzelfde protocol kan ook gebruikt worden voor transfectie van plasmide vectoren. Daarnaast kan de hier beschreven protocol eenvoudig worden uitgebreid om verschillende zoogdiercellijnen met kleine modificaties. Commerciële beschikbaarheid van MEA&#39;s met zowel vooraf gedefinieerde en aangepaste elektrode patronen te maken van deze techniek toegankelijk to meest onderzoekslaboratoria met basis celkweek apparatuur.
