Die Entdeckung von RNAi in Eukaryoten und der anschließenden Entwicklung von RNAi Mittel wie siRNA und shRNA, haben eine potente Verfahren zum Silencing spezifische Gene 1-8 für funktionelle Genomik und Therapeutika erreicht. Eine große Herausforderung bei RNAi basierenden Studien beteiligt ist die Lieferung von RNAi Agenten gezielt Zellen. Traditionelle non-viralen Techniken, wie Bulk-Elektroporation und chemische Transfektionsmethoden fehlt oft die notwendige räumliche Kontrolle über Lieferung und leisten schlechte Transfektionseffizienz 9-12. Jüngste Fortschritte in der chemischen Transfektionsmethoden wie kationische Lipide, kationische Polymere und Nanopartikel wurden in stark verbesserte Transfektionseffizienzen 13 geführt. Allerdings sind diese Techniken immer noch nicht präzise räumliche Kontrolle über Lieferzeiten, die immens Hochdurchsatz-Technologien, einzelne Zelle Studien und Untersuchung von Zell-Zell-Interaktionen können miniaturisierte Nutzen bieten. nt &quot;&gt; Neueste technologische Fortschritte in der Gentransfer haben High-Throughput-Transfektion von adhärenten Zellen 14-23, ein Großteil davon mikroskaligen Elektroporation verwenden aktiviert. Microscale Elektroporation bietet präzise räumlich-zeitliche Kontrolle über Lieferung (bis auf einzelne Zellen) und hat sich gezeigt, um hohe Wirkungsgrade 19, 24-26. Zusätzlich zu erreichen, müssen Elektroporation Ansätze erfordern eine längere Inkubationszeit (in der Regel 4 Stunden) mit siRNA und DNA-Komplexe wie nötig in der chemischen Basis Transfektionsmethoden und führen zu direkten Eingabe der nackte siRNA und DNA Moleküle in die Zelle Zytoplasma. Als Folge Genexpression kann bereits sechs Stunden nach der Transfektion 27 erreicht werden. Unser Labor hat zuvor die Verwendung von Mikroelektrodenarrays (MEA) für die ortsspezifische Transfektion in adhärenter Säugerzellkulturen 17-19 gezeigt. In der MEA Ansatz, erfolgt die Lieferung der genetischen Nutzlast über lokale Mikro-Maßstab electroporati erreichtauf der Zellen. Eine Anwendung des elektrischen Impulses an den ausgewählten Elektroden erzeugt lokale elektrische Feld, die Elektroporation von Zellen in der Region der stimulierten Elektroden führt. Die unabhängige Steuerung der Mikroelektroden stellt räumliche und zeitliche Steuerung über Transfektion und ermöglicht auch mehrere Transfektion basierte Experimente auf der gleichen Kultur durchgeführt werden Erhöhung des Durchsatzes und zur Verringerung experimentellen Kultur-zu-Kultur Variabilität. Hier beschreiben wir den Versuchsaufbau und das Protokoll für die gezielte Transfektion von adhärenten HeLa-Zellen mit einem fluoreszenzmarkierten verschlüsselten Sequenz siRNA mittels Elektroporation. Das gleiche Protokoll kann auch zur Transfektion von Plasmid-Vektoren verwendet werden. Zusätzlich kann das hier beschriebene Protokoll leicht an eine Vielzahl von Säugetier-Zelllinien mit geringfügigen Modifikationen verlängert werden. Kommerzielle Verfügbarkeit von MEAs sowohl mit vordefinierten und benutzerdefinierten Elektrode Muster machen diese Technik zugänglich to die meisten Forschungslabors mit grundlegenden Zellkultur Ausrüstung.
