ex vivo live imaging of single cell divisions in mouse neuroepithelium

Vi udviklede et system, der integrerer levende billeder af fluorescerende markører og dyrkning skiver af embryonale muse neuroepithelium. Vi tog fordel af eksisterende muselinjer til genetisk celleafstamning sporing: a tamoxifen-inducerbar Cre linje og en Cre reporter linje udtrykker DsRed upon Cre-medieret rekombination. Ved hjælp af et relativt lavt niveau af tamoxifen, var vi i stand til at inducere rekombination i et lille antal celler, tillader os at følge individuelle celledelinger.
