ex vivo live imaging of single cell divisions in mouse neuroepithelium

Wir entwickelten ein System, das Echtzeit-Bildgebung von fluoreszierenden Markern und Kultivierung von embryonalen Maus Scheiben Neuroepithel integriert. Wir nutzten vorhandene Mauslinien für genetische Zelllinie Tracing: eine Tamoxifen-induzierbare Cre Linie und einer Linie, die Cre-Reporter auf Cre-vermittelte Rekombination dsRed. Durch die Verwendung einer relativ niedrigen Tamoxifen, waren wir in der Lage, um eine Rekombination in einer kleinen Anzahl von Zellen zu induzieren, was uns erlaubt, einzelne Zellteilungen folgen. Darüber hinaus beobachtet man die transkriptionelle Reaktion auf Sonic Hedgehog (Shh) Signal mit einer Olig2-eGFP transgenen Linie 1-3 und man überwacht Bildung von Zilien mittels Infektion des kultivierten Scheibe mit Virus, das die Wimpern Marker SSTR3 4-GFP. Um das Bild Neuroepithel wir geerntet Embryonen bei E8.5, isoliert das Neuralrohr, montiert die neurale Scheibe in der richtigen Kulturbedingungen in die Kammer und Bildgebung durchgeführt Zeitraffer-konfokale Bildgebung. Unsere exLive vivo-Bildgebungsverfahren ermöglicht es uns, einzelne Zellteilungen zu verfolgen, um die relative Timing Hauptwimpern Bildung und Shh Antwort in einem physiologisch relevanten Weise zu beurteilen. Dieses Verfahren kann leicht angepasst werden, mit unterschiedlichen fluoreszierenden Markern und bietet das Feld die Werkzeuge, mit denen das Verhalten der Zelle in situ und in Echtzeit zu überwachen.

Hier führten wir ex vivo Echtzeit-Bildgebung von einzelnen Zellteilungen im E8.5 Maus Neuroepithel. Um einzelne Zellen markieren, induziert wir Cre-Rekombinase in einer Untergruppe von Zellen, die ein Cre-Reporter Linie, die bei der Rekombination 5,6 (3A) dsRed ausgedrückt.
