ex vivo live imaging of single cell divisions in mouse neuroepithelium

Vi har utviklet et system som integrerer direkte avbildning av fluorescerende markører og dyrking skiver av embryonale mus neuroepithelium. Vi tok nytte av eksisterende mus linjer for genetisk celle avstamning tracing: en tamoxifen-induserbar Cre linje og en Cre reporter linjen uttrykker DsRed på Cre-mediert rekombinasjon. Ved å bruke et relativt lavt nivå av tamoxifen, var vi i stand til å indusere rekombinasjon i et lite antall celler, tillater oss å følge de enkelte celledelinger. I tillegg observerte vi transcriptional svar på Sonic Hedgehog (Hysj) signal ved hjelp av en Olig2-eGFP transgene linjen 1-3 og vi overvåkes dannelsen av flimmerhårene ved å infisere den kultiverte skive med virus som uttrykker flimmerhårene markør, Sstr3-GFP fire. For å avbilde neuroepithelium, vi høstet embryoer på E8.5, isolert nevralrøret, montert det nevrale skive i riktig dyrking forhold til bildebehandling kammeret og utførte time-lapse confocal bildebehandling. Vår exvivo direkte avbildning metoden gjør oss i stand til å spore enkelt celledelinger for å vurdere den relative timing av primære cilier dannelse og Hysj respons i en fysiologisk relevant måte. Denne metoden kan enkelt tilpasses ved hjelp av ulike fluorescerende markører og gir feltet de verktøyene som brukes til å overvåke celle atferd in situ og i sanntid.

Her har vi utført ex vivo direkte avbildning av encellete divisjoner innenfor E8.5 mus neuroepithelium. Å merke individuelle celler, indusert vi Cre recombinase i en undergruppe av celler som inneholder en Cre reporter linje som uttrykt DsRed ved rekombinasjon 5,6 (figur 3A).
