Nosso sistema ex vivo nos permite observar diretamente divisões celulares individuais dentro do neuroepithelium em tempo real em desenvolvimento. ex vivo live imaging of single cell divisions in mouse neuroepithelium

Nós desenvolvemos um sistema que integra imagens ao vivo de marcadores fluorescentes e as fatias de cultura de neuroepithelium embrionário mouse. Aproveitamos mouse linhas existentes para a célula de linhagem genética traçado: a linha Cre tamoxifeno induzível e uma linha repórter Cre expressar dsRed sobre recombinação Cre-mediada. Ao utilizar um nível relativamente baixo de tamoxifeno, fomos capazes de induzir a recombinação em um pequeno número de células, que permite que se siga as divisões celulares individuais. Além disso, observou-se a resposta de transcrição para o Sonic Hedgehog (Shh) de sinalização usando um OLIG2-eGFP transgênico linha 1-3 e acompanhámos formação de cílios por infectar a fatia culta com vírus expressando o marcador de cílios, Sstr3-GFP 4. A fim de imagem neuroepithelium, colhemos embriões em E8.5, isolada do tubo neural, montado a fatia neural em condições de cultura adequadas para a câmara de imagens e realizada lapso de tempo de imagem confocal. Nosso exmétodo in vivo de imagens ao vivo nos permite traçar divisões celulares individuais para avaliar a temporização relativa de formação de cílios e resposta Shh de um modo fisiologicamente relevante. Este método pode ser facilmente adaptado por meio de marcadores fluorescentes distintas e fornece o campo com o qual as ferramentas para controlar o comportamento de células in situ e em tempo real.

Aqui realizamos ex vivo de imagens ao vivo de divisões celulares individuais dentro do E8.5 rato neuroepithelium.
