Nuestro sistema ex vivo nos permite observar directamente las divisiones de células individuales dentro del neuroepitelio en desarrollo en tiempo real. ex vivo live imaging of single cell divisions in mouse neuroepithelium

Hemos desarrollado un sistema que integra imágenes en vivo de los marcadores fluorescentes y las rebanadas de cultivo embrionario neuroepitelio del ratón. Nos aprovechamos de las actuales líneas de ratones de células linaje genético rastreo: una línea Cre tamoxifeno-inducible y una línea reportero Cre expresan dsRed por recombinación Cre-mediada. Mediante el uso de un nivel relativamente bajo de tamoxifeno, hemos sido capaces de inducir la recombinación en un pequeño número de células, lo que permite que sigamos divisiones celulares individuales. Además, se observó la respuesta transcripcional de la proteína Sonic Hedgehog (Shh) de señalización utilizando un Olig2-EGFP transgénicos línea 1-3 y que supervisó la formación de los cilios mediante la infección de la porción cultivada con virus que expresa el marcador de los cilios, SSTR3-GFP 4. Con el fin de imagen del neuroepitelio, que cosecharon embriones a E8.5, aislado del tubo neural, montado la rebanada neuronal en condiciones de cultivo adecuadas en la cámara de formación de imágenes y realiza de lapso de tiempo de imagen confocal. Nuestro exvivo método de imagen en directo nos permite trazar divisiones celulares individuales para evaluar el tiempo relativo de formación de los cilios primarios y la respuesta de Shh en forma fisiológicamente relevante. Este método se puede adaptar fácilmente el uso de marcadores fluorescentes distintos y proporciona el campo de las herramientas con las que monitorean el comportamiento de células in situ y en tiempo real.

Aquí hemos realizado ex vivo de imágenes en directo de las divisiones de células individuales dentro de la E8.5 neuroepitelio ratón. Para marcar las células individuales, se indujo la recombinasa Cre en un subconjunto de células que contienen una línea reportero Cre que expresaban DsRED tras la recombinación 5,6 (Figura 3A).
