Funktionelle Inaktivierung der Genexpression in Säugetierzellen ist für die Untersuchung der Beitrag eines Proteins von Interesse, um verschiedene Bahnen 1,2. Jedoch wird bedingten Knockdown der Genexpression in Fällen erforderlich, wenn konstitutive Knockdown nicht durch Zellen für einen langen Zeitraum 3-5 toleriert. Hier beschreiben wir ein Protokoll für die Herstellung von Zelllinien erlaubt bedingten Knockdown Untereinheiten des ACF Chromatin-Remodeling-Faktor. Diese Zelllinien erleichtern die Bestimmung des Beitrags von ACF, um die Induktion des Zelltodes durch das Adenovirus-Protein E4orf4 6. Sequenzen, die für kurze Haarnadel RNAs und für die Acf1 SNF2h Untereinheiten des ACF Chromatin-Remodeling-Faktor wurden neben einer Doxycyclin-induzierbaren Promotors in einem Plasmid auch ein Gen enthält, das für Neomycin-Resistenz-Gen kloniert. Neomycin-resistente Zellklone wurden in Gegenwart von G418 und getrennt ausgewählt. Die resultierenden Zelllinien wurden durch induzierteDoxycyclin-Behandlung, und einmal Acf1 oder SNF2h Expressionsniveaus wurden reduziert, wurden die Zellen mit einem Plasmid E4orf4 oder einem leeren Vektor transfiziert. Um die spezifische Wirkung der shRNA Konstrukte zu bestätigen, wurden Acf1 oder SNF2h Proteinniveaus zu WT Ebenen durch Kotransfektion mit einem Plasmid exprimieren Acf1 oder SNF2h die gegen das shRNA gemacht wurden durch Einführen stiller Mutationen wiederhergestellt. Die Fähigkeit von E4orf4 zum Zelltod in den verschiedenen Proben zu induzieren, wurde durch eine DAPI-Assay, in dem die Häufigkeit des Auftretens von Keimen mit apoptotischen Morphologien in der transfizierten Zellpopulation 7-9 gemessen wurde. Die hier beschriebene Protokoll kann zur Bestimmung der funktionellen Beitrag verschiedener Proteine, um die Induktion des Zelltods durch ihre Proteinpartner in Fällen verwendet werden, wenn konstitutive Knockdown-Zelle tödlich sein kann.
