<p> Hoewel de muis onderhuidse tumor modellen zijn eenvoudig vast te stellen en te controleren, is het duidelijk dat dit model niet kan repliceren de oorspronkelijke anatomische site van colorectale kanker. Door het verschil in micro-omgeving, hebben colorectale kankercellen groeien onder de huid is aangetoond dat hun fenotype te veranderen en bijna altijd geen vooruitgang en metastaseren <sup>1,2.</sup> In feite kan de tumor respons op de behandeling sterk variëren, afhankelijk van de vraag of kankercellen worden geïmplanteerd in een ectopis...

orthotopic mouse model of colorectal cancer

<p class="jove_content">Het traditionele subcutane tumor model is minder dan ideaal voor het bestuderen van colorectale kanker. Orthotope muismodellen van colorectale kanker, die kankercellen groeien in hun natuurlijke plaats kenmerk, repliceren de menselijke ziekte met high-fidelity. Twee technieken kunnen worden gebruikt om dit model vast te stellen. Beide technieken zijn vergelijkbaar en vereisen muis verdoving en laparotomie voor blootstelling van de blindedarm. Een techniek betreft injectie van een colorectale kanker celsuspensie in de blindedarmkeutels muur. Kankercellen worden eerst gekweekt in cultuur, geoogst wanneer subconfluent en voorbereid als een enkele celsuspensie. Een klein volume van de cellen wordt langzaam geïnjecteerd om lekkage te voorkomen. De andere techniek bestaat transplantatie van een stukje onderhuidse tumor op de blindedarm. Een muis met een eerder vastgestelde onderhuidse colorectale tumor is gedood en de tumor wordt verwijderd met behulp van steriele techniek. De tumor stuk is verdeeld in kleine stukjes voor transplantatie naar een andere muis. Voorafgaand aan de transplantatie, is de blindedarmkeutels muur lichtjes beschadigd tumorcellen infiltratie mogelijk te maken. De tijd om het ontwikkelen van primaire tumoren en levermetastasen zal variëren afhankelijk van de techniek, cellijn, en gebruikt muizensoorten. Dit orthotope muis model is handig voor het bestuderen van de natuurlijke progressie van colorectale kanker en testen van nieuwe therapeutische middelen tegen colorectale kanker.</p>

<p class="jove_content">Twee technieken kunnen worden gebruikt om dit model te stellen: injectie van een kankercel suspensie in de blindedarmkeutels muur of transplantatie van een stukje van subcutane tumor op de blindedarm.
