multi-analyte biochip (mab) based on all-solid-state ion-selective electrodes (assise) for physiological research

Lab-on-a-chip (LOC) toepassingen in milieu, biomedische, landbouw, biologische en ruimtevaart onderzoek vereisen een ion-selectieve elektrode (ISE) dat langdurige opslag kan weerstaan ​​in complexe biologische media 1-4. Een all-solid-state ion-selectieve-electrode (ASSISE) is vooral aantrekkelijk voor de bovengenoemde toepassingen. De elektrode moet de volgende gunstige eigenschappen: eenvoudig constructie, weinig onderhoud, en (potentiële) miniaturisatie, waardoor voor batchverwerking. Een microfabricated ASSISE bestemd voor het kwantificeren H +, Ca2 + en CO 3 2 - ionen werd geconstrueerd. Het bestaat uit een edel-metalen elektrodelaag (bijvoorbeeld Pt), een transductie-laag en een ion-selectief membraan (ISM) laag. De laagfuncties transductie de concentratie afhankelijke chemische potentiaal van de ion-selectieve membraan transduceren in een meetbaar elektrisch signaal. THij levensduur van een ASSISE blijkt afhankelijk te handhaven van de potentiaal op de geleidende laag / membraan-interface 5-7. Om de ASSISE werken levensduur te verlengen en daardoor stabiel potentials te handhaven op het grensvlak lagen, hebben we gebruik gemaakt van de geleidende polymeer (CP) poly (3,4-ethyleendioxythiofeen) (PEDOT) 7-9 in plaats van zilver / zilverchloride (Ag / AgCl) de transducer laag. We geconstrueerd de ASSISE in een lab-on-a-chip formaat, dat noemden we het multianalytmethoden biochip (MAB) (Figuur 1). Kalibraties testoplossingen aangetoond dat de MAB kan controleren pH (werkbereik pH 4-9), CO 3 2 - (gemeten van 0,01 mM - 1 mM), en Ca2 + (log-lineaire bereik 0,01 mM tot 1 mM). De MAB voor pH zorgt voor een bijna-Nernstian helling reactie na bijna een maand van opslag in algen medium. De carbonaat biochips tonen een potentiometrische vergelijkbaar met dat van een gebruikelijke ion-selectieve elektrode. Physioltief voor farmacologische metingen werden gebruikt om biologische activiteit van het modelsysteem, het microalga Chlorella vulgaris controleren. De MAB brengt een groot voordeel, veelzijdigheid en gemultiplexte analyt sensing mogelijkheden, waardoor het voor vele beperkt toezicht situaties op aarde en in de ruimte. Biochip ontwerp en experimentele methoden De biochip is 10 x 11 mm in afmeting en heeft 9 Assisen aangewezen als werkende elektroden (WES) en 5 Ag / AgCl referentie-elektrode (RE&#39;s). Elke werkende elektrode (WE) is 240 micrometer in diameter en is op gelijke afstand op 1,4 mm van de RE&#39;s, die zijn 480 micrometer in diameter. Deze elektroden zijn verbonden met elektrische contactvlakken met een afmeting van 0,5 mm x 0,5 mm. Het schema wordt getoond in figuur 2. Cyclische voltammetrie (CV) en galvanostatische depositie methoden worden gebruikt om de PEDOT films met een bioanalytische electropolymerizeal Systems Inc (BASI) C3 cel stand (figuur 3). De teller-ion voor het PEDOT film is afgestemd op de analyt ionen van interesse. Een PEDOT met poly (styreensulfonaat) tegenion (PEDOT / PSS) wordt gebruikt voor H + en CO 3 2 -, terwijl een met sulfaat (toegevoegd aan de oplossing als CaSO 4) wordt gebruikt voor Ca2 +. De elektrochemische eigenschappen van het PEDOT-gecoate WE wordt geanalyseerd met behulp van cv&#39;s in redox-actieve oplossing (dwz 2 mM kaliumferricyanide (K 3 Fe (CN) 6)). Op basis van het CV profiel werd Randles Sevcik-analyse gebruikt om het werkzame oppervlak 10 bepalen. Spin-coating bij 1500 tpm wordt gebruikt om gegoten ~ 2 micrometer dik ion-selectieve membranen (ISM) op het MAB werkende elektroden (WES). De MAB in een microfluïdische stroom-cel kamer gevuld met 150 ul volume algen-medium, de contactvlakken zijn elektrisch verbonden met de BASI systeem (Figure 4). De fotosynthetische activiteit van Chlorella vulgaris wordt toezicht gehouden in omgevingslicht en donkere omstandigheden.
