Vår &quot;LeafJ&quot; plugin gjør måling av petiole lengde semi-automatisk, øke gjennomstrømning nesten 6 ganger over manuell måling. leafj: an imagej plugin for semi-automated leaf shape measurement

Høy gjennomstrømning fenotyping (phenomics) er et kraftig verktøy for å knytte gener til deres funksjoner (se gjennomgang 1 og nyere eksempler 2-4). Bladene er primær fotosyntetiske orgel, og deres størrelse og form varierer developmentally og miljømessig innenfor en plante. For disse grunner studier på blad morfologi krever måling av flere parametre fra mange blader, som er best gjort av semi-automatiske phenomics verktøy 5,6. Baldakin skyggen er en viktig miljømessig cue som påvirker plante arkitektur og livshistorie, suiten av svarene er kollektivt kalt skyggen unngå syndrom (SAS) 7. Blant SAS responser, skygge indusert blad petiole tøyelighet og endringer i bladområde er spesielt anvendbare som indekser 8. Oppdatert, kan blad form programmer (f.eks SHAPE 9, LAMINA 10, 11 LeafAnalyzer, LEAFPROCESSOR 12) måle blad skisserer og kategorisere blad figurer, Men kan ikke sende petiole lengde. Mangel på store målesystemer av blad petioles har hemmet phenomics tilnærminger til SAS forskning. I denne artikkelen beskriver vi en nyutviklet ImageJ plugin, kalt LeafJ, som raskt kan måle petiole lengde og blad blad parametere av modellen anlegget Arabidopsis thaliana. For sporadisk blad som krevde manuell korrigering av petiole / blad blad grense vi brukte en touch-screen tablet. Videre blad cellen form og blad celle tall er viktige determinanter for blad størrelse 13. Atskilt fra LeafJ vi også presentere en protokoll for å bruke en touch-screen tablet for måling cellen form, område og størrelse. Vår blad egenskap målesystem er ikke begrenset til skygge-unngåelse forskning og vil akselerere blad fenotyping mange mutanter og screeningmetoder planter ved blad fenotyping.
