Inneren Organen wie Herz, Gehirn und Darm entwickeln Links-Rechts-(LR) Asymmetrien, die kritisch für ihre normale Funktion 1 sind. Motile Zilien sind bei der Festlegung LR Asymmetrie in Wirbeltierembryonen, einschließlich Maus, Frosch und Zebrafisch 2-6 beteiligt. Schicksal Mapping hat gezeigt, dass KV aus einer Gruppe von ~ 20-30 Zellen als dorsalen Vorläuferzellen (DFCs), die an der dorsalen Rand Blastoderm wandern während epiboly Stufen 8,9 bekannt abgeleitet ist. Während der frühen Somiten Stufen, um DFCs Cluster und differenzieren sich in Flimmerepithelzellen KV in der Schwanzknospe des Embryos 10,11 bilden. Die Fähigkeit, zu identifizieren und zu verfolgen DFCs-in Kombination mit optischen Transparenz und rasche Entwicklung des Zebrafisch-Embryos-make Zebrafisch KV ein hervorragendes Modell System LR Flimmerzellen studieren. Unter Ausnutzung dieser zytoplasmatischen Brücken, entwickelten wir ein Stadium-spezifische Injektion Strategie Morpholino Oligonucleotide liefernotides (MO) ausschließlich DFCs und Knockdown die Funktion eines gerichteten Gens in diesen Zellen 12. Diese Technik schafft chimären Embryos in dem Genfunktion in dem DFC / KV Linie entwickeln im Kontext eines Wildtyp-Embryos wird geklopft. Um asymmetrische Strömung in KV analysieren, injizieren wir fluoreszierenden Mikrokügelchen in den KV Lumen und aufzeichnen bead Bewegung mit Videomikroskopie 2. Fluidströmung einfach visualisiert und können durch die Verfolgung bead Verschiebung im Laufe der Zeit quantifiziert werden. Stufenspezifischen MO Injektionen stellen einen nützlichen Ansatz Genfunktion in bestimmten Kompartimenten des Embryos zu analysieren.
