PALM beelden bevatten informatie over molecule telt en posities binnen een cel, waardoor gedetailleerde analyse van de verdeling en plaatsing van doeleiwit moleculen die moeilijk te bereiken met andere middelen. super-resolution imaging of the bacterial division machinery

Bacteriële celdeling vereist de gecoördineerde verzameling van meer dan tien essentiële eiwitten op midcell 1,2. Centraal in dit proces is de vorming van een ringvormige suprastructuur (Z-ring) van de FtsZ eiwit op perceelsgrenzen 3,4. De Z-ring uit meerdere enkelstrengs FtsZ protofilamenten en begrijpen van de opstelling van de protofilamenten in de Z-ring inzicht in het mechanisme van Z-ringarmatuur en zijn functie als krachtgenerator 5,6. Deze informatie is moeilijk te realiseren vanwege huidige beperkingen in conventionele fluorescentie microscopie en elektronenmicroscopie. Conventionele fluorescentiemicroscopie staat is een hoge resolutie van de Z-ring door de diffractielimiet van licht (~ 200 nm) te verschaffen. Electron cryotomographic beeldvorming heeft gedetecteerd verspreid FtsZ protofilamenten in kleine C. crescentus cellen 7, maar is moeilijk toepasbaar op grotere cellen zoalsE. coli of B. subtilis. Hier de toepassing van een super-resolutie fluorescentie microscopie methode beschrijven, fotogeactiveerde Localization Microscopy (PALM) kwantitatief kenmerken de structurele organisatie van de E. coli Z-ring 8. PALM beeldvorming biedt zowel een hoge ruimtelijke resolutie (~ 35 nm) en specifieke etiketteringsvoorschriften om eenduidige identificatie van doeleiwitten mogelijk te maken. We gelabeld FtsZ de fotoactiveerbare fluorescerend eiwit mEos2, die van groene fluorescentie (excitatie = 488 nm) schakelt rode fluorescentie (excitatie = 561 nm) bij activering bij 405 nm 9. Tijdens een PALM experiment, worden enkele FtsZ-mEos2 moleculen stochastisch geactiveerd en de bijbehorende zwaartepunt posities van de individuele moleculen worden bepaald met &lt;20 nm precisie. Een super-resolutie afbeelding van de Z-ring wordt vervolgens gereconstrueerd door superpositie van het zwaartepunt posities van alle gedetecteerde FtsZ-mEos2 moleculen. &lt;p class = &quot;jove_content&quot;&gt; Met deze methode is dat de Z-ring een vaste breedte van ~ 100 nm heeft en bestaat uit een losse bundel FtsZ protofilamenten die met elkaar overlappen in drie dimensies. Deze gegevens fungeren als aanknopingspunten voor verder onderzoek van de celcyclus afhankelijke veranderingen van de Z-ring 10 en kan worden toegepast op andere eiwitten van belang.

In figuur 3Aiv een tweedimensionale superresolutie weergave van de Z-ring gegenereerd uit de PALM imaging hierboven beschreven werkwijze. Hieronder vatten we kwalitatieve en kwantitatieve informatie die kan worden verkregen van hen.
