Biologische activiteit wordt gecontroleerd door vergaderingen van eiwitten en andere biomoleculen die in onderling overleg handelen om cellulaire functies katalyseren. Sprekende voorbeelden zijn de machines die de erfelijke informatie in DNA in messenger RNA transcribeert. In mensen, meer dan 100 eiwitten samenkomen in een gedefinieerd en gereguleerd proces om genen transcriberen, die grote multiprotein complexen met 10 en meer subeenheden waaronder RNA polymerase II en algemene transcriptiefactoren zoals TFIID, TFIIH en anderen. 9 Andere voorbeelden zijn de ribosoom, bestaande uit vele eiwitten en RNA moleculen, die eiwitsynthese of de nucleaire porie complex is verantwoordelijk voor shuttling biomoleculen door de nucleaire envelop in eukaryoten katalyseert. Een gedetailleerde architectuur en biochemische dissectie van nagenoeg alle multicomponent machines in de cel is essentieel om de functie te begrijpen. Om dergelijke complexen toegankelijk te gedetailleerde structurele en functionele analyse maken vereist heterologe productie met behulp van recombinant techniques. Productie van recombinant eiwit had een grote impact op life science onderzoek. Vele eiwitten zijn recombinant geproduceerd en hun structuur en functie ontleed bij hoge resolutie. Structurele genomica programma&#39;s hebben gebruik gemaakt van de opheldering van de genomen van vele organismen aan het genproduct repertoire van gehele organismen in high-throughput mode (HT) te pakken. Duizenden eiwitstructuren zijn aldus bepaald. Tot op heden is de meest overvloedig gebruikt voor recombinant eiwitproductie geweest E. coli en vele expressiesystemen ontwikkeld en verfijnd door de jaren heterologe in deze gastheer. De plasmiden een overvloed aan functionaliteiten om eiwitproductie mogelijk in E. coli vullen hele catalogi van commerciële aanbieders. Echter, E. coli heeft bepaalde beperkingen waardoor het ongeschikt voor veel eukaryote proteïnen te produceren en in particulaire eiwitcomplexen met vele subeenheden. Daarom heeft eiwitproductie in eukaryotische gastheren steeds meer de werkwijze van keuze in de afgelopen jaren. Een bijzonder geschikt systeem eukaryote eiwitten is het baculovirus expressie-vectorsysteem (BEV) dat steunt op een recombinant baculovirus die de heterologe genen insecten celkweken gekweekt in het laboratorium infecteren. Het MultiBac systeem is een recent ontwikkelde BEV die vooral is aangepast voor de productie van eukaryotische eiwitcomplexen met vele subeenheden (Figuur 1). MultiBac werd voor het eerst geïntroduceerd in 2004. 13 Sinds de introductie, heeft MultiBac voortdurend verfijnd en gestroomlijnd om behandeling te vereenvoudigen, te verbeteren doelproteïne kwaliteit en in het algemeen maken van het systeem toegankelijk is voor niet-gespecialiseerde gebruikers door het ontwerpen van efficiënte standard operating procedures (SOP&#39;s). 4 MultiBac is geïmplementeerd in veel laboratoria in de wereld, in academia en industrie. Aan het EMBL in Grenoble, werden transnationale toegang programma&#39;s opgezet door de Europese Commissie om deskundig opleiding op het MultiBac platform voor wetenschappers die wilden deze productie systeem te gebruiken voor het bevorderen van hun onderzoek. De structuur en functie van vele eiwit-complexen die tot dusverre niet toegankelijk waren opgehelderd door monsters geproduceerd MultiBac. 4 Hierna worden de essentiële stappen van MultiBac productie samengevat in protocollen als in werking op MultiBac faciliteit in EMBL Grenoble.

Proteomics onderzoek bleek de indrukwekkende complexiteit van eukaryote proteomen in ongekend detail. MultiBac is een BEV die in het bijzonder is ontworpen voor de productie van eukaryotische eiwitcomplexen die veel subeenheden bevatten. 4 Een essentiële voorwaarde voor efficiënte productie van eiwitten en hun complexen stabielere protocollen voor alle bij een expressie experiment die ideaal kan worden geïmplementeerd als stappen standard operating procedures (SOP&#39;s) en volgde ook door niet-gespecialiseerde gebruikers betrekkelijk gemakkelijk.
