Proteomics Forschung enthüllt die beeindruckende Komplexität von eukaryotischen Proteom in bisher unerreichter Genauigkeit. Die Baculovirusexpressionsvektor System (BEVS) hat sich als besonders nützlich für die Herstellung von eukaryotischen Proteinen, deren Aktivität oft auf posttranslationale Prozessierung, dass andere häufig verwendete Ausdruck Systeme oft kannnicht unterstützt. 3 BEVS mit einem rekombinanten Baculovirus, in die das Gen von Interesse eingeführt wird, um Insekten Zellkulturen, die wiederum das Protein der Wahl infizieren wurde. MultiBac eine BEVS, die besonders wurde für die Herstellung von eukaryontischen Protein-Komplexe, die viele Untereinheiten enthalten zugeschnitten. 4 Eine wesentliche Voraussetzung für die Herstellung von Proteinen und deren Komplexe sind stabil Protokolle für alle Schritte in einem Ausdruck Experiment, das sich hervorragend als implementiert werden beteiligt Standard Operating Procedures (SOPs) und auch von Nicht-Spezialisten Nutzer vergleichsweise einfach gefolgt. Die MultiBac Plattform am European Molecular Biology Laboratory (EMBL) verwendet SOPs für alle Schritte in einer Multiproteinkomplex Ausdruck Experiment beteiligt, ab Einsetzen der Gene in einer konstruierten baculovirale Genom für die heterologe Proteinproduktion Immobilien zum kleinen Analyse des Proteins optimiert Exemplare produziert. 5-8 Die Plattformwird in einer Open-Access-Modus bei EMBL Grenoble installiert und unterstützt viele Wissenschaftler aus Hochschule und Industrie, um Protein-Komplex Forschungsprojekte beschleunigen.
