Die Bestimmung der Anzahl von Plasmiden pro Nanopartikel komplexiert ist eine wirksame Nanopartikel-Genabgabe Strategien, insbesondere zum Zusammenwirken Bereitstellung von mehreren Plasmiden zur selben Zelle Target gestalten, wie es oft in Stammzelle Umprogrammierung Studien 12 erforderlich. In einer anderen Studie verwendet Collins et al eine Strömung Teilchenfluß-Bildanalyse Technik zu studieren (Lys) 16-enthaltende Peptid / DNA-Komplexen;. Jedoch ihre Methode nur auswerten größeren, Mikrometergröße Partikel 16. Somit haben wir kürzlich entwickelte eine neuartige flexible Assay, um die Anzahl von Plasmiden pro Nanopartikel 11 quantifizieren.

Nicht-virale Gentransfer unter Verwendung von polymeren Nanopartikeln als attraktiver Ansatz für Gentherapie zur Behandlung genetischer Krankheiten 1 und als Technologie für die regenerative Medizin 2 entstanden. Im Gegensatz zu Viren, die erhebliche Sicherheitsprobleme haben, können Polymernanopartikel gestalten als nicht-toxisch, nicht-immunogenen, nicht-mutagen, leichter zu synthetisieren, chemisch vielseitig für die Beförderung von Fracht größeren Nukleinsäure und biologisch abbaubare und / oder ökologisch anspricht werden. Kationischen Polymeren mit negativ geladenen DNA über elektrostatische Wechselwirkung auf Komplexe in der Größenordnung von 100 nm, die üblicherweise polymere Nanoteilchen bezeichnet bilden selbst zusammen. Beispiele von Biomaterialien verwendet werden nanoskalige polykationischen Genabgabe Nanopartikel bilden, umfassen Polylysin, Polyphosphoester, Poly (Amidoamine) s und Polyethylenimin (PEI), welches ein nicht-abbaubaren off-the-shelf kationisches Polymer ist üblicherweise für Nukleinsäurelieferung 1,3 verwendet. Zusätzlich wird ein Hochdurchsatz-96-Well-Platte für Transfektions-Assay schnelle Screening der Transfektion Wirksamkeit von polymeren Nanopartikeln vorgestellt. Bei diesem Protokoll, Poly (beta-Aminoesters) s (PBAEs) als Modell Polymeren und menschlichen retinalen Endothelzellen (HRECs) verwendet werden als mo benutztdel menschlichen Zellen. Dieses Protokoll kann leicht angepasst werden, um jegliche polymere Nanoteilchen und eine beliebige Zellart von Interesse in einer Multiwellplatte Format auswerten.

Abbildung 1 zeigt eine fluoreszenzmikroskopische Aufnahme eines Beispiels für eine erfolgreiche Transfektion HRECs mit der EGFP-Plasmid.
