Das hier beschriebene Protokoll erlaubt die Analyse von Zellzyklus, bezogen Zellgröße und relative Zellzahl in Live Drosophila Gewebe bei verschiedenen Entwicklungsstadien. Durchflusszytometrie können für Messungen der Lebensfähigkeit der Zellen, bezogen Zellgröße, DNA-Gehalt und fluoreszierende Protein-Expression in lebenden Zellpopulationen verwendet werden. Durch die Replikation von Kern-DNA während der S-Phase, über DNA-Gehalt in einer Population von Zellen, die den relativen Prozentsätze der verschiedenen Phasen des Zellzyklus 1-3 abzuleiten. Diese Methode hat sich zu einem Eckpfeiler der Zellzyklus-Analyse in Modellsystemen von Hefe bis zu den Säugetieren. Die Fruchtfliege Drosophila melanogaster hat sich ein hervorragendes Modell für genetische Analysen in vivo Regulation des Zellzyklus. Die umfangreiche genetische Werkzeuge zur Verfügung, in den Fliegen ermöglichen elegant gewebespezifische und zeitlich geregelt Manipulationen Zellzyklusregulatoren zusammen mit in vivo Fluoreszenz-Protein-basierten Linie Tracing 4-6. Durchflusszytometrie wurde verwendet, um DNA-Gehalt in einer Reihe von Drosophila Zelltypen zu untersuchen, einschließlich endoreplicating Zellen und kultivierten Mitosezellen 7,8. Diese werden in der Regel nur in Sorter (dh BD FACS Vanta gefundenge, BD FACSAria) oder teure multicolor benchtop Systemen (zB BD LSR), in der Regel erfordern die Unterstützung durch institutionelle Flow Core Facilities. Wir haben die Hoechst-basiertes Protokoll modifiziert, um eine neue Live-Zell-DNA-Farbstoff von Invitrogen, Vybrant DyeCycle Violet verwenden. Dieser Farbstoff ist kompatibel mit einem violetten 405 nm Laser, häufiger in kleineren Tisch-Analysatoren und in der kleinen Selbstversorger-Tisch-Analysator, der Attune Acoustic Fokussierung Cytometer. Hier präsentieren wir Ihnen eine detaillierte Protokoll für Zellzyklus-Analyse, mit Zelltyp Zellgröße, Zahl und Abstammung Analyse in einer Vielzahl von Geweben Drosophila während der verschiedenen Phasen der Entwicklung mit DyeCycle Violet und die Attune gekoppelt werden kann. Dieses Protokoll erweitert die Anzahl der Zytometern geeignet für eine solche Analyse mit Drosophila Gewebe und gibt Beispiele, wie diese Art von Live-Zellzyklus-Analyse kann für weitere Gewebetypen und Entwicklungsstadien geändert werden.

Durchflusszytometrie wurde in großem Umfang verwendet, um Informationen über DNA-Gehalt in einer Population von Zellen zu erhalten, um die relative Prozentsätze in den verschiedenen Phasen des Zellzyklus zu schließen.
