Inademing is de meest waarschijnlijke blootstellingsroute voor mensen werken met aerosolizable gemanipuleerde nano-materialen (ENM). Aan nanodeeltjes inhalatie toxicologisch onderzoek goed uit te voeren, de aërosolen in een kamer waar de proefdieren moeten: 1) een constante concentratie gehandhaafd op een gewenst niveau voor de gehele periode de blootstelling; 2) een homogene samenstelling vrij van verontreinigingen, en 3) een stabiele grootteverdeling met een geometrische gemiddelde diameter van &lt;200 nm en een geometrische standaardafwijking σ g &lt;2.5 5. Het genereren van aërosolen met nanodeeltjes is heel uitdagend, omdat nanodeeltjes gemakkelijk agglomeraat. Dit is grotendeels te wijten aan sterke krachten tussen de deeltjes en de vorming van grote fractalstructuren in tientallen of honderden micron 6, die moeilijk te worden opgebroken worden. Een aantal gemeenschappelijke aërosols, waaronder vernevelaars, wervelbedden, Venturi afzuigers en de Wright stof voer, weopnieuw getest, maar niemand was in staat om nanodeeltjes aërosolen die aan alle criteria voldoen 5 produceren. Een hele lichaam aërosol inhalatie blootstelling systeem nanodeeltjes werd gefabriceerd, gevalideerd en gebruikt voor nano-TiO2 inhalatie toxicologische studies. Kritische componenten: 1) nieuwe nano-TiO2 Aërosolgenerator; 2) 0,5 m 3 whole-body exposure kamer inademing, en 3) de monitor en controlesysteem. Nano-TiO2 aërosolen gegenereerd bulk droge nano-TiO2 poeder (primaire diameter van 21 nm, stortdichtheid van 3,8 g / cm 3) werden in de blootstellingskamer bij een stroomsnelheid van 90 LPM (10,8 luchtverversing / hr) . Deeltjesgrootteverdeling en massaconcentratie profielen werden continu gemeten met een scanning mobiliteit deeltje Sizer (SMPS), en een elektrische lage druk impactor (ELPI). De aërosolmassa concentratie (C) werd gravimetrisch geverifieerd (mg / m 3). De massa (M) Van de verzamelde deeltjes werd bepaald als M = (M na pre M), waarbij M en M pre Post zijn massa van het filter vóór en na de monstername (mg). De kamerdruk, temperatuur, relatieve vochtigheid (RH), O2 en CO2 concentraties werden gecontroleerd en continu gecontroleerd. Nano-TiO2 aërosolen op Nuclepore filters opgevangen werden geanalyseerd met een scanning elektronenmicroscoop (SEM) en energie-dispersieve X-stralen (EDX) analyses. Samengevat, melden wij dat de nano-deeltjes aërosolen opgewekt en geleverd aan onze belichtingskamer hebben: 1) steady massaconcentratie; 2) homogene samenstelling vrij van verontreinigingen; 3) stabiele deeltjesgrootteverdelingen met een count-mediaan aerodynamische diameter van 157 nm tijdens aërosol generatie. Dit systeem betrouwbaar en herhaaldelijk testen creëert sferen die beroeps, ENM aerosol milieu of woning te simuleren.

Een studie inademing meestal gaat het handhaven van een proefdier in een bekende en constante testomgeving terwijl het blootstellen van het proefdier naar een gedefinieerde concentratie van een testmateriaal 8,9.
