Dit is een experimenteel muizenmodel van cutane wondgenezing. Een belangrijk kenmerk van dit model is het gebruik van siliconen spalken voorkomen wondcontractie zodat re-epithelialisatie en nieuwvorming weefsel optreden, zodat het op het proces dat plaatsvindt in mensen. Dit model is veelzijdig en kan worden gebruikt om wondheling in zowel fysiologische en pathofysiologische (bijvoorbeeld diabetes mellitus) waarden te beoordelen. Verstoorde wondgenezing is verantwoordelijk voor aanzienlijke morbiditeit en mortaliteit, dit geldt met name voor patiënten van diabetes mellitus 1,2. Bij mensen wondgenezing een continuüm van processen, waarbij er een aanzienlijke overlapping 3. Onmiddellijk na verwonding, inflammatoire processen worden geïnitieerd. Ontstekingscellen vrij factoren die de processen van celproliferatie, migratie en angiogenese stimuleren. Na re-epithelialisatie en nieuwvorming weefsel er een fase van remodeling dat zowel apoptose en reorganisatie van matrix eiwitten zoals collageen meebrengt. De complexiteit van de wondheling kan momenteel niet worden gerepliceerd in vitro en dit is het gebruik van diermodellen. Tot op heden zijn wondhelende studies beperkt tot grote diermodellen, zoals varkens, dat de genezingsprocessen gelijkwaardig zijn en vergelijkbaar met de mens. Echter, het gebruik van grote animals deze studies kan moeilijk zijn huis en zijn niet altijd praktisch 4. Het laboratorium muis vertegenwoordigt een economisch diermodel dat gemakkelijk genetisch kunnen worden gemanipuleerd mechanistische onderzoek 5-7. Echter, muizen wonden verschillend op de mens, vooral door het proces van contractie 8. Dit is gedeeltelijk te wijten aan een uitgebreide onderhuidse dwarsgestreepte spierlaag genaamd de panniculus carnosus dat is grotendeels afwezig in de mens. In muizen, deze spierlaag laat de huid los van de diepere spieren bewegen en is verantwoordelijk voor de snelle samentrekking van de huid volgt verwonding. Om deze beperking te overwinnen, kunnen murine wondgenezing aangepast aan menselijke wondgenezing repliceren door middel van een splint (figuur 1) 8,9. In deze video laten we zien de gespalkt muizen wondmodel dat wondcontractie elimineert en meer overeenstemt met de menselijke processen van re-epithelializatie en de nieuwe formatie weefsel. In dit model twee volle dikte uitsnijdingen dat de panniculus carnosus zijn worden op het dorsum, een aan elke kant van de middellijn van de muis. Een silicone splint is geplaatst rond de wond met behulp van lijm en de spalk vervolgens vastgezet met onderbroken hechtingen. Elke muis fungeert als zijn eigen controle, met een wond die behandeld en het andere voertuig controle, waardoor het aantal dieren verminderen. Na topische toepassingen, is een transparante afsluitend verband aangebracht. Het verband kan worden verwijderd indien nodig verder topische toepassingen en / of meting van het wondgebied 10,11. Bij de voltooiing van experimenten wondsluiting morfologische architectuur en de mate van neovascularisatie kan worden beoordeeld door immunohistochemie. Deze economische en gemakkelijk te voeren model kan ook worden gebruikt om wondheling in het kader van diabetes mellitus of andere pathophysiologies beoordelen.

Wondgenezing en reparatie zijn de meest complexe biologische processen die zich in het menselijk leven. Na verwonding, verschillende biologische routes geactiveerd worden. Verstoorde wondheling, die voorkomt bij diabetici bijvoorbeeld kan leiden tot ernstige ongunstige resultaten zoals amputatie. Er is dus een toenemende impuls aan nieuwe middelen die wondheling bevorderen. Het testen van deze is beperkt tot grote diermodellen zoals varkens, die vaak onpraktisch. Muizen vertegenwoordigen de ideale preclinical model aangezien ze zuinig en vatbaar voor genetische manipulatie, die zorgt voor mechanistische onderzoek. Echter, wondgenezing in een muis is fundamenteel verschillend van die van de mens als het voornamelijk via contractie. Onze muizenmodel ondervangt dit doordat een spalk rond de wond. Door spalken de wond, is het herstelproces dan afhankelijk epithelisatie, cellulaire proliferatie en angiogenese, die nauw de spiegelbiologische processen van menselijke wondgenezing. Terwijl die consistentie en verzorging, dit muizenmodel niet om ingewikkelde chirurgische technieken en maakt de robuuste testen van veelbelovende middelen die kunnen bijvoorbeeld angiogenese bevorderen of ontstekingsremmende. Bovendien, elke muis fungeert als eigen controle als twee wonden worden bereid, zodat de toepassing van zowel de testverbinding en controle voertuig op hetzelfde dier. Concluderend demonstreren we een praktische, eenvoudig te leren en robuuste uitvoering van wondgenezing, vergelijkbaar met die van mensen.
