Hoewel een aantal eerdere studies chronische infusie van glucose (bijv. 10-15) en lipiden (bijvoorbeeld 16,17) in knaagdieren zover wij weten hebben gebruikt de gecombineerde infusie van beide brandstoffen slechts gemeld bij muizen 18. De chronische infusie hier gepresenteerde model biedt verschillende voordelen voor de effecten op nutriënt overmaat studie over diverse biologische functies in ratten. Ten eerste, niet leidt genetisch obese knaagdieren en gemeenschappeli...

Chronisch verhoogde niveaus van glucose en lipiden in het bloed zijn voorgesteld bij te dragen aan de pathogenese van type 2 diabetes door het veranderen van de functie van verschillende organen betrokken zijn bij de handhaving van glucosehomeostase waaronder, maar niet beperkt tot beta-cel (beoordeeld in 1). De glucotoxiciteit hypothese stelt dat chronische hyperglycemie verergert de beta-cel gebrek dat aanleiding gaf tot hyperglycemie in de eerste plaats, waardoor een vicieuze cirkel en het bijdragen aan de verslechtering van glucose controle bij type 2 diabetespatiënten. Ook de glucolipotoxicity hypothese stelt dat gelijktijdige verhoging van glucose en lipiden, zoals vaak waargenomen bij type 2 diabetes, schadelijk zijn voor de beta cellen. Ontcijferen van de cellulaire en moleculaire mechanismen van de schadelijke effecten van chronisch verhoogde voedingsstoffen op beta-cel functie is de sleutel tot het understanding de pathogenese van type 2 diabetes 1. Daartoe is een groot aantal studies de mechanismen van chronische nutriënt overmaat ex vivo bestudeerd in geïsoleerde eilandjes van Langerhans in vitro of in klonale, insuline uitscheidende cellijnen. Echter, de vertaling van de resultaten verkregen in deze modelsystemen het hele organisme complex, met name omdat de concentratie van vetzuren in gekweekte cellen of eilandjes zelden overeen met de circulerende niveaus in de nabijheid van de beta-cellen in vivo 2. Aan de andere kant hebben de mechanismen van beta-cellen falen in reactie op voedingsstoffen overtollige onderzocht in diermodellen van diabetes, zoals wordt geïllustreerd door de Zucker Diabetic Fatty rat 3,4, de gerbil Psammomys obesus 5 en vetrijk dieet- gevoed muis 6. Deze modellen worden echter gekenmerkt door een intrinsieke metabole afwijkingen en niet lastig om manipulaties van bloedglucoseen / of lipiden in een meer gecontroleerde en minder chronische omstandigheden. Om veranderen circulerende niveaus van voedingsstoffen in een tijdsbestek van dagen anders normale dieren, hebben we een chronische infusie model in normale ratten waardoor we de effecten van lipiden en glucose onderzoeken, alleen of in combinatie ontwikkeld op fysiologische parameters en functie 7,8.

Chronische blootstelling aan grote hoeveelheden nutriënten wordt gepostuleerd dat de functie van verschillende organen en weefsels beïnvloeden en bijdragen aan de ontwikkeling van de vele complicaties geassocieerd met obesitas en het metabool syndroom, waaronder type 2 diabetes. De procedure bestaat uit catheterizing rechts halsader en de linker halsslagader onder algehele narcose, waarbij een 7-daagse herstelperiode, verbinden de katheters om de pompen met een draaibare en contragewicht systeem dat de dieren zich vrij kunnen bewegen in de kooi, en infunderen glucose en / of Intralipid (een soja-olie-emulsie die een mengsel van ongeveer 80% unsaturated/20% verzadigde vetzuren genereert wanneer toegediend met heparine) gedurende 72 uur. Dit model biedt verschillende Advantages, inclusief de mogelijkheid om fijn te moduleren het streefniveau van circulerende glucose en vetzuren, de mogelijkheid om samen te bezielen farmacologische verbindingen, en het relatief korte tijdsbestek in tegenstelling tot voedings-modellen. Het kan worden gebruikt om de mechanismen van nutriënt-geïnduceerde disfunctie onderzocht in diverse organen en de doeltreffendheid van geneesmiddelen in deze context te testen.

Uit van een reeks van 42 ratten die een operatie ondergingen, werden 5 ratten verloren tijdens de postoperatieve periode en 1 rat werd verloren tijdens de infusie, wat neerkomt op een totale succespercentage van 86%. Het gemiddelde lichaamsgewicht van de 37 ratten die uiteindelijk werden toegediend was 608 ± 5 g voor de operatie en 588 ± 6 g aan het begin van de infusie (gemiddelde ± SE, n = 37, p &lt;0,0001 gepaarde t-test).
