Magnetic Resonance Imaging (MRI) gilt als das bildgebende Verfahren der Wahl für die Demonstration von infektiösen Synovitis aufgrund seiner hohen räumlichen Auflösung und Weichgewebe Gegensatz sein. Signal Veränderungen im MRT bei Arthritis niedrig T1 und T2 hohen Signal spiegelt die verstärkte Präsenz von extrazellulären Wassergehalt und eine deutliche Verbesserung nach Gadolinium-basierte Kontrastmittel Verabreichung beobachtet, im Einklang mit den Ergebnissen der histologisch erhöhte Durchblutung infolge Vasodilatation und Angiogenese 1. Dennoch ist MRI oft nicht Auflösung der Infektion während einer antibiotischen Therapie zeigen als persistente Verbesserung in den Gelenken von Patienten mit klinisch und biologisch geheilt septische Arthritis durch Persistenz vergrößerten extrazellulären Raum (Granulationsgewebe, fibrotische Narbe) 2 beobachtet werden. Neben extrazellulären Veränderungen, Entzündungszellen einschließlich einer intensiven Rekrutierung von Makrophagen massiv in die infiltrierenSynovia betroffen. Makrophagen spielen eine wichtige Rolle sowohl in der akuten und chronischen Phase einer bakteriellen Infektion 3. Sie induzieren die entzündliche Reaktion erforderlich ist, um pathogene Mikroorganismen zu beseitigen, und koordinieren Entzündung Auflösung, wie Makrophagen-induzierte Entzündung besteht, solange nekrotische Gewebe nicht entfernt werden, Verhindern Reparatur bis 4 beginnen. So kann die Reduktion von Makrophageninfiltration in infizierten Synovia nach effizienten Therapie ein zuverlässiger Indikator für die Auflösung der Infektion sein. Cellular Imaging ist in der Lage, spezifische zelluläre Infiltration im pathologischen Gewebe zu demonstrieren. Spezifische Makrophagen MR-Bildgebung durch gezielte Kontrastmittel wurde vielfach untersucht und hat gezeigt, seine Fähigkeit zu gemeinsamen Entzündung oder Infektion 5-7 demonstrieren. Nach intravenöser Injektion sind solche gezielte Kontrastmittel wie USPIO (ultrakleine superparamagnetische Eisenoxide) Partikel von Phagozyten wie Makrophagen aufgenommen undaufgrund ihrer magnetischen Eigenschaften zu induzieren Signaländerungen in Geweben präsentiert Makrophageninfiltration 8. Die Längs-Follow-up von denen MR-Signals ändert Therapie ermöglicht eine nicht-invasive in vivo Evaluierung Makrophageninfiltration, und eine Reduktion der USPIO beladenen Makrophagen-ähnlichen Signal Änderungen würden Therapieerfolg zu demonstrieren. Der Zweck dieses Berichts ist es bis heute, wie Makrophagen MR-Bildgebung durchführen, um nicht-invasiv überwacht septische Arthritis durch den Nachweis der Reduktion von Makrophageninfiltration innerhalb der Synovia.

Makrophagen sind Schlüssel-Zellen in der Initiierung, die Entwicklung und die Regulierung der Entzündungsreaktion auf bakterielle Infektion. Makrophagen sind intensiv und zunehmend in septischen Gelenke von den frühen Phasen der Infektion rekrutiert und die Infiltration soll einmal zurückbilden effiziente Beseitigung der Erreger erreicht wird. Die Fähigkeit, in vivo Makrophagenaktivität in einer infizierten gemeinsamen identifizieren können daher zwei Hauptanwendungen: Früherkennung der akuten Synovitis und Überwachung der Therapie. In vivo-invasive Erfassung von Makrophagen können mit Magnet-Resonanz-Bildgebung mit Eisen-Nanopartikeln wie ultrakleine superparamagnetische Eisenoxid (USPIO) durchgeführt werden. Nach intravaskulären oder intraartikuläre Verabreichung USPIO speziell von aktivierten Makrophagen und aufgrund ihrer magnetischen Eigenschaften phagozytiert, induzieren Signaländerungen in Geweben präsentiert Makrophageninfiltration. Ein quantitative Auswertung des infiltrieren möglich, da der Bereich mit einem Signalverlust (Anzahl dunkler Bildpunkte) auf Gradientenecho MR-Bildern beobachtet Teilchen nach Injektion mit der Menge an Eisen in dem Gewebe korreliert und daher gibt die Anzahl der USPIO-beladenen Zellen. Wir stellen Ihnen hier ein Protokoll zu Makrophagen Bildgebung mit USPIO-enhanced MR-Bildgebung in einem Tiermodell der septische Arthritis, so führen Sie einen Anfangs-und Längs-invasive in vivo Evaluierung von Makrophagen Infiltration und eine Bewertung der Therapie Aktion.
