De voor-en nadelen van de TED-methode zijn uitgebreid besproken in recente publicaties 34,35. Vergeleken met andere werkwijzen die hierboven beschreven TED omzeilt het probleem te verstoren en perfuseren van de cellen. Bovendien, twee aspecten moeten worden benadrukt. Om de fysiologische calcium reacties van de ER op te lossen, hebben we een nieuwe strategie om de vangst van synthetische calcium-gevoelige kleurstoffen in de ER verbeteren ontwikkeld. De methode kan de directe, niet-verstorende real-time monitoring van de vrije ER calcium in aanwezigheid van extracellulair calcium. Functie en signalering van ER Calcium Calcium signalen zijn te vinden in verschillende celtypen, bijvoorbeeld spier-cellen, neuronen en gliacellen en hun functies variëren van bemiddelen spiercontractie om een betrokkenheid bij synaptische transmissie in leren en geheugen 1,2. Veranderingen in de concentratie vrij calcium van groot wetenschappelijk belang omdat calcium is betrokken bij de regulatie van gentranscriptie, celproliferatie, neuronale prikkelbaarheid, celdood en andere cel signalerende gebeurtenissen 1-7. Al deze cellulaire calcium signalen functioneel verbonden en bijdragen tot intracellulaire calciumwinkel dynamiek 8-10. Een gemeenschappelijk kenmerk van alle calcium signalen is de stroom van calcium tussen de extracellulaire ruimte, het cytosol en organellen, vooral de ER en mitochondria. Hierdoor dynamische veranderingen in calciumconcentratie in deze organellen, die worden waargenomen door verschillende signaleringscomponenten. In het algemeen is de calciumconcentratie in het ER varieert tussen 100-800 uM, in het cytosol de calciumconcentratie dicht bij 100 nM en in de extracellulaire ruimte is de concentratie ongeveer 1-2 mM. Dienovereenkomstig is er een grote chemische motor voor calcium stromen naar het cytosol 2,9,10. De meest onderzochte ER-afgeleide calcium signalen afhankelijk van de stimulatie van G-eiwit gekoppelde receptoren (GPCR) die vervolgens activeren fosfolipase C (PLC). PLC op zijn beurt produceert inositol 1,4,5-trifosfaat (IP-3) 1. Na binding van IP3 zijn receptor (IP 3-Rec, figuur 1) in de ER-membraan, worden calciumionen vrij uit het ER lumen. Historisch gezien, IP 3-gemedieerde calcium vrijlating uit de ER was eerste - hoewel indirect - gemeten in acinar alvleeskliercellen door Streb et al. in 1983 11.. Deze publicatie voorgesteld voor het eerst een signalerende cascade met acetylcholine, fosfolipase C en IP3. Deze manier van calcium release algemeen aangeduid IP3-geïnduceerde calciumafgifte (IiCR) (figuur 1). Kinase-afhankelijke activering van fosfolipase Cy door receptor tyrosine kinases banden de actie van groeifactoren en neurotrofe factoren aan ER calcium signalering na IP 3 elevatie 12. Naast IiCR, kan calcium hoogte worden gemedieerd door ionotrope calciumingang, bijvoorbeeld via spanningsafhankelijke calciumkanalen (Ca V) en vervolgens calcium geïnduceerde calcium afgifte (CICR) door re ryanodineceptors (RyR). IiCR en CKP zijn fysiologisch gekoppeld aan winkel-bediende calciumingang (SOCE). SOCE omvat de werking van STIM (stroma interactie molecule), een sensor voor ER calcium release. STIM is aangetoond extracellulair calcium ingang stimuleren door transient receptor potential kanalen (Trp) 13 Orai calciumkanalen 14 en zelfs spanningsafhankelijke calciumkanalen 15 (figuur 1). Verlies van ER calcium wordt dynamisch gered door de werking van de sarco-endoplasmatisch reticulum calcium ATPase (SERCA), die actief pompen calcium terug in het ER. Het blokkeren van de SERCA met drugs zoals thapsigargin onthult een continu verlies van ER calcium aan de cytosolische compartiment. Deze ER calcium &quot;lek&quot; wordt veroorzaakt door ER intramembrane porie complexen zoals het Sec61-eiwitcomplex 16,17 (Figuur 1). In 1998 Berridge publiceerde een model, de &quot;neuron binnen een neuron model&quot;, which suggereert een principe fysiologische rol van de ER in het integreren van neuronale calcium 5. Dit model beschouwt het bestaan ​​van een continu ER membraan systeem vormen een intracellulair &quot;imago&quot; van de neuronale plasmamembraan 5. Dan kunnen hun som effecten op de prikkelbaarheid van het neuron, regulatie van gen transcriptie of integratie van signaleringscascades hebben. Dus de ER ondersteunt de integratie van calcium signalen. Een voorwaarde voor dit concept is de continuïteit van de ER in een cel, die is geclaimd door verscheidene studies die bewezen althans somato-dendritic gebieden en de korte afstand axonale projecties 19-21. Of sprake is van ER continuïteit binnen lange axonale projecties is een kwestie van debat. Strategieën om de stroom van vrij calcium via ER membraan meten Calcium signalen worden meestal gevolgd in het cytosol 22,23. Derhalve kan niet gemakkelijk worden onderscheiden of Ca 2 + stroomt in het cytosol van extracellulaire of intracellulaire opslagplaatsen 6,24. Om deze beperking te omzeilen, hebben methodologische strategieën voor directe ER calcium beeldvorming ontwikkeld. Samenvattend worden de volgende strategieën: (1) ER-gerichte genetisch gemanipuleerde eiwit-indicatoren 25-27 eiwit gebaseerde lage affiniteit Ca2 + indicatoren de bioluminescent eiwit GFP aequorine of in combinatie met een calcium gevoelige eiwit.. Deze genetisch gemanipuleerde Ca 2 +-indicatoren (GECIS) kangericht naar de ER met behulp van een signaalpeptide en actief blijven in het ER met een retentietijd en retrieval motief. Gemeenschappelijke ER Ca 2 +-indicatoren te baseren op de Cameleon principe en zijn de Cameleon YC4.3 26,28; Cameleon split YC7.3ER 29, en Cameleon D1 30. (2) Direct esterase-based dye laden van AM-ester lage affiniteit Ca 2 +-indicatoren 31,32. AM-derivaten van de indicator kleurstoffen (Mag-fura2-AM, Mag-Fluo4-AM of Fluo5N-AM) passeren de biologische membranen in een lipofiele, calcium-ongevoelige toestand. (3) AM-ester loading in combinatie met plasma membraan permeabilisatie 32. Overgebleven cytosolische Ca 2 +-indicator wordt verwijderd door plasma-membraan permeabilisatie met kleine hoeveelheden van een &quot;mild&quot; detergens (bijvoorbeeld saponine) in een kunstmatige intracellulaire buffer. Een cel wordt eerst geladen met een lage affiniteit Ca 2 +-indicator (bijv. Mag-fura2- AM, ratiometrisch, UV-licht). TED vereist overexpressie van een ER gerichte muis carboxylesterase (CES2) (Figuur 2), hetgeen wordt bereikt door expressieconstructen. Cellen die een recombinant CES-construct worden geïncubeerd met het AM-ester vorm van calcium inindicator kleurstof (Fluo5N-AM, figuur 2). De werkwijze is bijzonder nuttig voor ER calcium-afgifte te onderzoeken via de IiCR-wegen bijvoorbeeld via metabotrope, purinerge-of glutamaatreceptoren 8,34 en ER calcium uitputting visualiseren via &quot;lek kanalen&quot; direct, bijvoorbeeld na blokkade van de SERCA 17 , 34. Onze ervaring de lage affiniteit Ca 2 +-indicator Fluo5N-AM is momenteel de best beschikbare indicator om te gebruiken met de TED kleurstof laden strategie. Fluo5N-AM heeft een lage affiniteit voor Ca 2 + (dissociatieconstante K D ~ 90 uM, figuur 2), is vrijwel non-tl in zijn AM-vorm, maar biedt een hoge fluorescentie-emissie bij calcium. binding 8,34. De Fluo5N/Ca 2 + complex geëxciteerd kan worden met een standaard lichtbron van ~ 490 nm, wat overeenkomt met standaard kleurstoffen zoals FITC, Alexa 488 of eGFP. Cytosolische calcium signalen nauwelijks een concentratie in het lage uM range bereiken en worden derhalve nauwelijks gedetecteerd door Fluo5N in de cytosol 35. Om TED verbeteren, werden verschillende recombinante vector constructen ontwikkeld (figuur 3). Oorspronkelijk TED vectoren op basis van de coderende sequentie van CES2 (Refseq toegangsnummer NM_145603, CES2c) en best TED werking wordt waargenomen met stabiele expressie van CES constructen. Nieuwe TED vectoren drukken een kernelement van CES2 gefuseerd met de rode fluorescentie eiwit TagRFP-T2 36. Deze vectoren hebben het voordeel dat zij kunnen worden geïdentificeerd getransduceerde cellen en de rode fluorescentie als een interne controle voor de normalisatie van veranderingen in Fluo5N/Ca 2 + fluorescentie. De rode fluorescentie biedt ook de mogelijkheid om de structurele verdeling van de ER en veranderingen in ER processen onder de stimulatiecondities visualiseren.

Visualisatie van calciumdynamiek is belangrijk om de rol van calcium in celfysiologie begrijpen. Aan calcium dynamiek te onderzoeken, hebben synthetische fluorescerende Ca2 + indicatoren populair geworden. Hier laten we TED (= gericht esterase-geïnduceerde loading dye), een werkwijze voor de afgifte van Ca2 + verbetering indicator kleurstoffen in het ER lumen van verschillende celtypes. Tot op heden werd TED in cellijnen, gliacellen en neuronen in vitro. TED bases op efficiënte, recombinant richten van een hoge carboxylesterase activiteit om het ER lumen met behulp van vector-constructies die express carboxylesterasen (CES). De meest recente TED vectoren bevatten een kernelement van CES2 gefuseerd met een rood fluorescerend eiwit, waardoor gelijktijdig twee-kleuren beeldvorming. De dynamiek van de vrije calcium in het ER worden afgebeeld in een kleur, terwijl de desbetreffende ER structuur verschijnt in het rood. Aan het begin van de procedure worden cellen getransduceerd met een lentivirus. Vervolgens, de geïnfecteerde cellen eenopnieuw gezaaid op dekglaasjes om eindelijk live cell imaging mogelijk. Vervolgens worden levende cellen geïncubeerd met de acetoxymethylester (AM-ester) vorm van lage-affiniteit Ca 2 +-indicatoren, bijvoorbeeld Fluo5N-AM, Mag-Fluo4-AM, of Mag-fura2-AM. De esterase activiteit in de ER klieft uit hydrofobe zijketens van de AM-vorm van de Ca 2 +-indicator en een hydrofiele fluorescerende kleurstof / Ca 2 + complex wordt gevormd en opgesloten in het ER lumen. Na kleurstof laden, worden de cellen geanalyseerd op een omgekeerde confocale laser scanning microscoop. Cellen zijn continu geperfuseerd met Ringer-achtige oplossingen en de ER calciumdynamiek direct gevisualiseerd door time-lapse imaging. Calcium afgifte uit het ER wordt geïdentificeerd door een afname in fluorescentie-intensiteit in de gebieden van belang, terwijl het bijvullen van de ER calcium opslag veroorzaakt een toename in fluorescentie-intensiteit. Tenslotte wordt de verandering in fluorescentie-intensiteit in de tijd bepaald door berekening van AF / F 0.

Dit protocol voorziet in een niet-verstorende benadering voor directe beeldvorming van gratis ER calcium. Lage affiniteit synthetische Ca2 + indicatoren worden vrijgegeven en gevangen in het ER lumen met behulp van een ER gerichte recombinante esterase enzymactiviteit.
