We ontwikkelden een embryo isolatieprotocol dat snel doeltreffende en kan gemakkelijk worden overgedragen naar andere laboratoria. De hier beschreven apparatuur bestaat uit een omgekeerde microscoop, een micromanipulator, glas microcapillairen, een verticale gloeidraad trekker en een microinjector (figuur 3A). De embryo van bloeiende planten wordt omgeven door het endosperm, een voedingsweefsel afgeleid van een tweede fertilisatiegebeurtenis. Zowel embryo en endosperm zijn omgeven door verschillende cellagen van de zaadhuid. Gezamenlijk vormen deze weefsels een zaad dat in de vrucht te ontwikkelen. Zo zijn weefsel-en cel-specifieke analyses van Arabidopsis embryo&#39;s sterk aangetast vanwege hun ontoegankelijkheid. Toch embryo&#39;s in de late-bolvormige of latere stadia zijn relatief goed vatbaar voor handmatige dissectie met behulp van fijne wolfraam naalden onder de stereomicroscoop, of door lichte druk op het zaad met behulp van een tang om ze te halen. Dergelijke technieken zijn met succes toegepast transcriptoom of epigenoom profilering analyses zoals microarray hybridisatie bisulfietsequencing of RNA sequentie (bijvoorbeeld 1-3). In tegenstelling, studies van embryo&#39;s in de zygote tot begin bolvormige stadium blijven technisch uitdagend. Tot op heden slechts een paar studies hebben reported transcriptoomanalyses op jonge embryo&#39;s met behulp van laser-capture microdissectie (LCM) van embryonale weefsels van vaste zaad hoofdstukken 4 of manuele extractie van individuele embryo vanuit zaden met behulp van fijne instrumenten 5. Echter, LCM apparatuur niet algemeen beschikbaar en handmatige extractie embryo in een vroeg stadium is tijdrovend en vereist uitstekende dissectie vaardigheden die niet makkelijk overdraagbaar. Naast genoom-brede analyses, in situ genexpressie analyses zijn ook moeilijk te voeren op jonge hele-mount embryo van Arabidopsis. Tot op zekere hoogte kunnen jonge embryo op objectglaasjes worden vrijgegeven door lichte druk op de zaden en gebruikt voor reporter assays of proteindetectie door immunokleuring (zie bijvoorbeeld 6,7). Deze techniek is echter niet mogelijk high-throughput embryo isolatie, waardoor kwantitatieve analyse belemmeren. Daarom is een efficiënt en snel ontwikkeld protocolvoor vroege embryo isolatie van Arabidopsis zaden die is eenvoudig op te zetten, gemakkelijk overdraagbaar, en geschikt voor een verscheidenheid van downstream toepassingen. Het basisprincipe is om voorzichtig te verpletteren zaden - ontleed van jonge siliques in een Eppendorf buisje met een plastic stamper in een geschikte isolatie buffer. Het extract wordt in druppels op een multi-well glijbaan en gescreend op de aanwezigheid van vrijgekomen embryo&#39;s in de gewenste fase met behulp van een omgekeerde microscoop. Embryo&#39;s worden verzameld met behulp van een glazen microcapillaire bevestigd aan een microinjector of een standaard laboratorium pipet. Voor moleculaire toepassingen, worden embryo&#39;s tweemaal gewassen door herhaalde introductie in druppels van nieuwe isolatie buffer voor het overzetten naar de bestemming buffer in een minimaal volume. Voor cytologische toepassingen (reporter assays, immunokleuring, FISH), kunnen wasstappen worden weggelaten. De methode biedt verschillende voordelen: (i) het levert 25-40 embryo ~ 45 min voor cytologische appcaties of 3-4 uur gedurende moleculaire toepassingen (waaronder de wasstappen), (ii) het mogelijk isoleren van specifieke embryonale stadia, (iii) het kan gemakkelijk worden overgedragen aan andere personen laboratoria vanwege de eenvoudige installatie, (iv) vereist betaalbaar Voorzieningen voor basale configuratie die vatbaar is voor upgrades, en (v) werd met succes gebruikt voor verschillende stroomafwaartse toepassingen zoals RNA sequencing 8, genspecifieke DNA-methylatie analyse 9 reporter assays (10 en Raissig et al.., in prep.) en VIS (J. Jaenisch, U. Grossniklaus, C. Baroux ongepubliceerde, zie figuur 5).

In bloeiende planten, het embryo zich ontwikkelt binnen een voedende weefsel - het endosperm - omringd door de moederlijke zaad integumenten (of zaadhuid). Als gevolg daarvan, is de isolatie van de plant embryo&#39;s in een vroeg stadium (1 cel tot bolvormige stadium) technisch uitdagende vanwege hun relatieve ontoegankelijkheid. Efficiënte handmatige dissectie in de aanloopfase wordt sterk belemmerd door de kleine omvang van jonge Arabidopsis zaden en de kleverigheid van het embryo aan het omringende weefsel. Hier beschrijven we een werkwijze die de efficiënte isolatie van jonge Arabidopsis embryo maakt, bevatten tot 40 embryo&#39;s in 1 uur tot 4 uur, afhankelijk van de stroomafwaartse toepassing. Embryo&#39;s worden uitgebracht in een isolement buffer met iets breken 250-750 zaden met een plastic stamper in een Eppendorf buis. Een glazen microcapillaire bevestigd aan ofwel een standaard laboratorium pipet (via een rubberen buis) of een hydraulisch gestuurde microinjector wordt gebruikt om embryo&#39;s te verzamelen van druppeltjes plaatsd op een multi-well slide op een omgekeerde lichtmicroscoop. De technische vaardigheden die nodig zijn eenvoudig en gemakkelijk overdraagbaar, en de basis-setup is dure apparatuur nodig. Verzamelde embryo&#39;s zijn geschikt voor verschillende stroomafwaartse toepassingen zoals RT-PCR, RNA sequentie, DNA methylatie analyse, fluorescentie in situ hybridisatie (FISH), immunokleuring en reporter assays.

Onze embryo isolatieprocedure (figuur 1) maakt isolatie van maximaal 40 embryo&#39;s in 4 uur als wassingen worden uitgevoerd, bijvoorbeeld voor moleculaire toepassingen, of in minder dan een uur als wast zijn weggelaten, bijvoorbeeld voor cytologische toepassingen.
