Cryo-electron tomography (cryoET) is een krachtige imaging techniek die driedimensionale (3D) visualisatie van cellen en weefsels mogelijk maakt en geeft inzicht in de organisatie van de inheemse organellen en cellulaire structuren op moleculaire resolutie in een dicht-bij fysiologische toestand 1. Echter, de inherent lage contrast van ongekleurde bevroren gehydrateerde monster, gecombineerd met hun stralingsgevoeligheid, bemoeilijkt gebieden plaats binnen een cel lokaliseren en vervolgens uitvoeren van de tilt series succes zonder beschadiging van het doelgebied. Om deze problemen te overwinnen, een correlatieve benadering die licht en elektronenmicroscopie combineert noodzakelijk. Bijzonderheden gemarkeerd door fluorescentielabelling worden geïdentificeerd en gelokaliseerd door fluorescentie microscopie, en vervolgens de coördinaten worden overgedragen aan de elektronenmicroscoop voor het ingaan van hoge resolutie 3D structurele gegevens. Deze correlatieve methode helpt het lokaliseren van de doelgroep eenReas van belang zijn worden aangepakt. Door de beperking van monsterdikte met cryoEM (&lt;300 nm), nog alleen de randgebieden van de cel geschikt voor 3D structuuranalyse door CryoET. Verdere vermindering van de dikte van bevroren gehydrateerd monsters door glasvocht snijden 8 of door cryo-focused ion beam (FIB) frezen 9 zou het vermogen van correlatieve beeldvorming breiden. Voorheen werden correlatieve methoden voornamelijk gebruikt om cryoET data acquisitie voor grote en statische constructies 10-13 vergemakkelijken. In deze studies zijn cryo-fasen geïmplementeerd om cryoEM roosters accepteren en passen op ofwel een rechtopstaande microscoop of een omgekeerde microscoop 10,11,14. Hoewel schakelende grids lijkt vrij eenvoudig in hun ontwerpen, zijn er extra stappen die overdracht van de EM rooster, waardoor de kans dat het rooster kan worden vervormd, beschadigd en vervuild. We hebben onlangs aangetoond een technische vooruitgang incorrelatieve microscopie die ons in staat stelt om direct te visualiseren dynamische gebeurtenissen die van nature moeilijk te vangen, zoals HIV-1 en gastheercel interactie in een vroeg stadium van de infectie 15. We bereikte dit door het ontwerpen en implementeren van een cryo-lichtmicroscopie monstertafel dat een cartridge systeem om het monster schade door raster handling minimaliseren aanpast, waardoor het gemakkelijker correlatie. Ons ontwerp omvat een geïntegreerd exemplaar patroonhouder, waardoor beide cryo-licht en cryo-elektronenmicroscopie worden uitgevoerd, achtereenvolgens, op hetzelfde monster houder, zonder monster overdracht, waardoor het stroomlijnen van het correlatieve proces. Daarnaast hebben we eveneens uitgevoerd een nauwkeurige en betrouwbare correlatie procedure met fluorescerende latex kralen als markeerders.

Cryo-electron tomography (cryoET) maakt het mogelijk 3D-visualisatie van cellulaire structuren op moleculaire resolutie in een close-to-fysiologische toestand 1. Echter, directe visualisatie van individuele virale complexen in hun gastheer cellulaire omgeving met cryoET 2 is uitdagend, gezien de lage frequentie en de dynamische aard van virale binnenkomst, met name in het geval van HIV-1. In deze video artikel, bieden we gedetailleerde methoden en protocollen voor structurele onderzoek van HIV-1 en gastheer-cel interacties met behulp van 3D correlatieve high-speed Live-cell imaging en hoge-resolutie cryoET structurele analyse. HeLa-cellen geïnfecteerd met HIV-1 deeltjes werden eerst gekenmerkt door confocale microscopie van levende cellen, en het gebied dat dezelfde virale deeltje werd vervolgens geanalyseerd met cryo-electron tomografie voor 3D structurele details.
