Het netvlies is een bijna transparante neuraal weefsel langs de achterkant van het oog. Veel studies gebruiken het netvlies als het model om de eerste stappen in de verwerking van visuele en mechanismen van synaptische signalering te onderzoeken. Aangezien de retinale netwerk van het-mount preparaat intact blijft na sectie, vormt dit een ideaal systeem synaptische interacties te bestuderen als fysiologische reacties zijn vergelijkbaar met de in vivo omstandigheden. Dus, met behulp van een geïsoleerde retina de eigenschappen van zijn neuronen kunnen worden bestudeerd met behulp van patch clamp technieken (voor reviews over de techniek, zie 6,9,13). Identificatie van de precieze bijdrage van specifieke circuits en neurotransmitters ganglion cel respons echter meestal belemmerd als farmacologische middelen werken op verschillende sites. Fysiologische reacties van retinale neuronen aan het licht, de natuurlijke stimulus, kan worden opgenomen met glazen pipetten gevuld met intracellulaire vloeistof. Met behulp van patch clamp technieken, neuronale reacties op licht stimulatie kan worden opgenomen als membraanpotentiaal fluctuaties (stroomtang) of als stromingen (voltage clamp). Door die de membraanpotentiaal op verschillende voltages en uitvoering a posteriori geleiding analyse is het mogelijk remmende en prikkelende synaptische inputs isoleren 5,12. Dit soort experimenten worden uitgevoerd in normale baden medium en in de aanwezigheid van verschillende farmacologische middelen om de bijdrage van verschillende neurotransmitters en receptoren neuronale reacties isoleren. Een schat aan studies van vele laboratoria gekarakteriseerd de afhankelijkheid van stekelige output en prikkelende en remmende ingangen op stimulus eigenschappen zoals grootte, contrast, ruimtelijke en temporele frequenties, richting, oriëntatie en andere stimulus variabelen. Hoewel deze experimentele benaderingen geven informatie over de relatie tussen de spike output en synaptische inputs als functie van stimulus eigenschappen,interpretatie van de bijdrage van specifieke celtypen en hun synaptische inputs naar cel exciteerbaarheid niet eenvoudig. Dit komt door het feit dat zowel typische prikkelende en remmende inputs variëren stimulus eigenschappen en dus is het niet mogelijk om het precieze effect dat veranderingen in een van deze parameters moet op neuronale spiking beoordelen. Een alternatieve benadering om deze beperkingen te omzeilen is voor het uitvoeren van dynamische clamp opnames, die een kritische evaluatie van de bijdrage van de individuele synaptische inputs naar stekelige uitgang toestaan. De dynamische clamp techniek maakt directe injectie van stroom in de cel en de hoeveelheid stroom geïnjecteerd in een bepaald afhankelijk van de opgenomen membraanpotentiaal destijds 1,2,3 (zie voor 7,14). Het is een gemodificeerde stroomklem set-up waarbij een real-time, snelle feedback interactie tussen de cel onder opname en de apparatuur bestaande uit gespecialiseerde hardware, softwareen een computer bereikt. De hoeveelheid stroom geïnjecteerd in de cel wordt dienovereenkomstig berekend. Vandaar dat het voordeel van deze methode is dat de cellen kunnen worden gestimuleerd met verschillende combinaties geleidingstijd golfvormen en de reactie zal de activatie van receptoren die synaptische inputs bemiddelen nabootsen. Bijvoorbeeld, vergelijking van de reactie op injectie van prikkelende en remmende geleidingen voor een kleine vlek met de reactie op injectie van prikkelende geleiding voor een kleine vlek alleen informatie over het effect van remming op de celrespons. Ook andere combinaties van fysiologisch opgenomen conductances collocatie kan worden geïnjecteerd om te onthullen hoe stimulus-afhankelijke veranderingen in prikkelende en / of remmende conductances invloed spike output. In onze studie is de dynamische clamp techniek gebruikt om het effect van de relatieve amplitude en tijdstip van synaptische inputs op de vuurgedrag van retinale ganglion cellen vertonen. Diverse conductantiesverkregen fysiologische experimenten controle omstandigheden of in aanwezigheid van farmacologische middelen werden gebruikt als input. Daarnaast zijn kunstmatige geleidingen gebaseerd op alpha functies ook gebruikt om te onderzoeken hoe synaptische inputs worden geïntegreerd door neuronen. Dit is dus een veelzijdige techniek die het mogelijk maakt verschillende soorten geleiding gegenereerd ofwel fysiologisch, farmacologisch of rekenkundig worden geïnjecteerd in dezelfde ganglion cel, zodat de respons op deze ingangen worden.
