Pluripotente stamceller har potensial til å differensiere spontant inn i CMS, riktignok med lav effektivitet og høy variasjon blant linjene. Utvikling av nye metoder for igangsetting har derfor fokusert på effektivisering av prosessen og beveger seg mot mer definerte protokoller. Historisk har etterforskningen av de genetiske og molekylære mekanismer som driver menneskelig utvikling og sykdom vært basert på produksjon av genmodifiserte dyremodeller. Imidlertid tallrike humane fenotyper ikke klarer å være vellykket replikeres i mus, hovedsakelig på grunn av de biologiske forskjeller som eksisterer mellom de to artene. På den annen side, tilgang på humant vev kan være begrenset og ofte ikke tillate nok materiale til å bli oppnådd for inngående eksperimentelle studier. Avledning av humane embryonale stamceller (ESC) første 5 og banebrytende oppdagelse av induserte pluripotente stamceller (iPS) celler ved Yamanaka og Thomson 6,7 senere syntes å tilby løsninger: Disse cellene har evnen til å vokse på ubestemt tid og potensial til å gi stige til alle celle derivater av de tre bakterie lag, herunder det CMS. De har derfor tillate utviklingen av in vitro modeller som letter etterforskningen av menneskelige genetiske sykdommer og mekanismer for utvikling. I kraft av denne karakteristiske, kan iPS celler også overcome problemer knyttet til immunforsvaret avvisning og etiske problemstillinger åtte. For disse grunner, selv om ESCs fortsatt representerer gullstandarden innen stamcelleforskningen biologi, er forskere over hele verden nå beveger seg mer mot iPS celle teknologi. iPS celler blir nå ansatt for å modellere menneskelige sykdommer in vitro for ulike forhold, inkludert medfødte kardiovaskulære lidelser 9,10. Nylig har anvendelsen av iPS celle sykdom modellering utført for monogene hjertesykdommer (dvs. lang QT syndromer, catecholaminergic polymorf ventrikulær takykardi) og sykdomstrekk der kardiale defekter er en del av en kompleks fenotype (dvs. Leopard og Timothy syndromer, dilatert kardiomyopati). Disse rapportene bekreftet at pasient-spesifikke iPS celler som skiller seg inn i CMS vise lignende fenotypiske og funksjonelle egenskaper som sykdommen in vivo. Men deter fortsatt mange utfordringer for å forbedre effekten av å indusere iPS celler i hjertets avstamning. Spontan generasjon av CMS fra menneskelige ESCs via aggregatdannelse kalt embryoid organer (EBS) har vist seg vellykket 17. Siden denne oppdagelsen, har mange andre metoder blitt foreslått. For å undersøke hendelsene kjøring hjerte utvikling og for å bestemme de molekylære mekanismene som fører til hjerteinfarkt sykdommer hos mennesker, er det viktig først å generere funksjonelle menneskelige cardiomyocytes (CMS). Bruken av disse celler i medisiner og toksikologiske studier ville også være meget gunstig, slik at nye farmakologiske molekyler for behandling av hjertesykdommer som skal valideres pre-klinisk på celler av human opprinnelse. I våre studier genererte vi CMS fra flere iPS cellelinjer.
